| Objective:By comparing attention deficit hyperactivity disorder model SHR rats and their normal controls WKY rats hypothalamus,hippocampal tissue morphology,hypothalamus CRH,GR,MR,hippocampal GR,MR protein expression differences,to observe the pathological changes of hippocampal-HPA in SHR rats,and on this basis to observe the effect of compound Longmu Guizhi Decoction on the protein expression of hypothalamus CRH,GR,MR and hippocampal GR,MR in SHR rats,to explore its treatment of ADHD by regulating hippocampal-HPA axis Mechanism of action.Methods:1.Taking young SHR rats as ADHD model group and WKY rats as control group,HE staining was used to observe the cell morphology of the hypothalamus and hippocampus of the two groups;immunohistochemistry was used to detect the protein expression of CRH,GR,MR in hypothalamus and GR,MR in hippocampus of the two groups.2.Young SHR rats were randomly divided into three groups: normal saline group,methylphenidate hydrochloride group(2mg· kg-1),Longmu Guizhi Decoction group(30g·kg-1).The protein expression levels of CRH,GR,MR in hypothalamus and GR,MR in hippocampus of the three groups were detected by immunohistochemistry after the corresponding drugs were given to the stomach for 14 days.Results:1.There was no significant difference in the morphology of hypothalamus and hippocampus between SHR rats in model group and WKY rats in control group,but the protein expression level of CRH in hypothalamus of SHR rats in model group was lower than that of WKY rats in control group(P < 0.05),while the protein expression level of GR and MR in hypothalamus,hippocampus was higher than that of WKY rats in control group(P < 0.05).2.The protein expression level of CRH,GR,MR in hypothalamus and GR,MR in hippocampus of rats in Longmu Guizhi Decoction group and methylphenidate group were higher than that in normal saline group(P < 0.05),and the protein expression level of GR in hippocampus of rats in Longmu Guizhi Decoction group was higher than that in methylphenidate group(P < 0.05).Conclusion:1.The pathogenesis of ADHD may be related to the low protein expression of CRH in hypothalamus and the high protein expression of GR and MR in hypothalamus and hippocampus.2.Longmu Guizhi Decoction can improve the function of hippocampal-HPA axis by up regulating the protein expression of CRH,GR,MR in hypothalamus and GR,MR in hippocampus,which may be one of the mechanisms of Longmu Guizhi Decoction in the treatment of ADHD. |
