Objective: To study the expression of gene and protein of Brunton’s tyrosine kinase(Btk)in clinical samples of chronic periodontitis by collecting clinical samples of chronic periodontitis,so as to provide reference for further study of the role of Btk in chronic periodontitis and the treatment of chronic periodontitis.Methods: the clinical samples of chronic periodontitis were collected and the expression of Btk gene and protein was detected by real-time fluorescence quantitative PCR and Western blot.(1)Clinical sample collectionFrom January 2018 to June 2019,30 patients between 21 and 59 years old were selected for treatment in our hospital.15 patients who needed periodontal surgery were in the experimental group,and 15 patients who needed flap extraction but had healthy periodontal were in the control group.Inform the patient of the purpose and content of this study,and sign with the informed consent of the patient.The above research was approved by the ethics committee of our institute.According to the requirements of the 4th edition of Periodontology published by the people’s Health Press and the classification standard of periodontal diseases of the International Symposium on periodontal diseases in 1999,the clinical examination indexes were recorded.The inclusion criteria of patients in the experimental group were: periodontal pocket and formation(pocket depth > 5mm),Cal > 3mm,GI ≥ 2,PLI ≥ 2,BOP(+);imaging showed that alveolar bone had absorption,alveolar bone absorption > 1 / 3 root length.The inclusion criteria of the healthy control group were: the gingival tissue of the third molars with impacted teeth flap extraction,Cal = 0;(2)GI = 0,PLI = 0,BOP(-);imaging showed no alveolar bone absorption;no other oral diseases,and no anti-inflammatory drugs were taken in the past month.(2)The expression of Btk gene was detected by real-time fluorescent quantitative PCR and Western blot.12 experimental groups and 12 healthy control groups were randomly selected to carry out real-time fluorescent quantitative PCR experiment;the remaining 3 experimental groups and 3 healthy control groups were carried out western blot experiment.The above gingival tissue was completely abraded,RNA was extracted from the sample tissue,RNA concentration was measured,RNA was reverse transcripted into c DNA,and the expression of Btk in m RNA level in gingival tissue was detected;Processing samples according to Western blot experimental operation requirements,glue making,the determination of protein concentration and protein denaturation,SDS-PAGE electrophoresis,the analysis of the odvalue and molecular weight of Btk bands in the experimental group and the healthy control group,and the detection of the expression of Btk at the protein level.All experimental results were collected and sorted out,and statistical analysis was carried out.result:1.Collection of clinical samplesThere were 15 patients in the experimental group,including 8 males and 7 females,aged from 23 to 59 years old,and 15 in the healthy control group,including 9 males and6 females,aged from 21 to 49 years old.2.Real time fluorescent quantitative PCR test resultsThe results of real-time fluorescent quantitative PCR showed that the expression of Btk in the gingival tissue of the experimental group and the healthy control group was higher than that of the healthy control group(P < 0.05).3.Western blot test resultsThe results of Western blot showed that the expression of Btk in gingival tissue of the experimental group and the healthy control group was higher than that of the healthy control group(P < 0.05).Conclusion: the expression of Btk in human chronic periodontitis suggests that Btk may participate in the disease process of chronic periodontitis,but its specific function in chronic periodontitis needs further study.
