| Objective:Discussed iron caulis dendrobii on ovarian rats osteoporosis bone protection element(osteoporogeterin,OPG)-nuclear factor kappa B ligand receptor activation factors predominate(receptor activator of the nf-kappa B ligand,RANKL)-nuclear factor kappa B receptors predominate activation factor(receptor activator of the nf-kappa B,RANK)axis of the impact of cell signaling pathways,So as to explore the tin caulis dendrobii prevention and treatment of Postmenopausal Osteoporosis(Postmenopausal Osteoporosis,PMOP)mechanism,as to provide theoretical basis for the development and utilization of iron caulis dendrobii.Methods:(1)Forty-eight 3-month old female Sprague-Free(SPF)SD rats weighing 200±10g were randomly divided into 36 rats in the model group,12 rats in the sham group,ovary removal in the model group,and sham group.After modeling,4 rats died in modeling group and 1 rat died in sham operation group.After 8 weeks,in the sham operation group,3 bone specimens were randomly selected from each model group to observe the bone structure,and the bone density of the lumbar body was measured by dual-energy X-ray absorptiometry.Test whether the mold is made successfully;After successful modeling,the remaining 29 rats in the modeling group were randomly divided into 10 rats in the high-dose group,10 rats in the low-dose group,9 rats in the OVX group(castrated rats)and 9 rats in the sham operation group.(2)The OVX group and the sham operation group were given the same volume of purified water by gavage at different doses(1.092g/mL in the low dose group and 4.368g/mL in the high dose group with the dose of 10ml/kg body weight(equivalent to the equivalent dose for clinical use in adults).The rats in each group were given the drug by gavage once a day,and the rats in each group were given the drug by gavage once a day for 6 consecutive days each week.Weigh yourself once a week and adjust the dose accordingly.All experimental animals were fed under the same conditions except for differences in gavage.At the weekly time nodes of 4(high-dose group,low-dose group,OVX group,and sham operation grroup)and 8(high-dose group,low-dose group,AND OVX group,and sham operation group),rats were randomly selected from each group for dual-energy X-ray bone mineral density(BMD)measurement.Blood was collected from abdominal aorta.Enzyme linked immunosorbent assay(ELISA)was used to detect the levels of Estradiol(E2)and OPG/RANKL/RANK.After 8 weeks of administration,the lumbar vertebrae were observed by electron microscope for comparative analysis.Results:①After 8 weeks of modeling,compared with the sham operation group,X-ray film and bone mineral density index analysis of the modeling group indicated that the model was established successfully.②Results of bone metabolism Ca and P showed that compared with OVX group,serum Ca and P levels were significantly increased in the low and high dose groups at 4 weeks and 8 weeks after administration,with statistically significant differences(P<0.05).③Results of lumbar bone mineral density showed that compared with OVX group,lumbar bone mineral density was significantly increased in the low and high dose groups at 4 weeks,8 weeks,and the difference was statistically significant(P<0.05).④Serum E2 and OPG/RANK/RANKL results showed that compared with OVX group,the serum OPG content in low and high dose groups at 4 weeks and 8 weeks of administration was significantly increased(P<0.05).Compared with OVX group,for 4 weeks,8 weeks,serum content of RANK high dose group were significantly lower,the difference was statistically significant(P<0.05),for 8 weeks the content of serum RANK low dose group was obviously lower,the difference was statistically significant(P<0.05),4 weeks for serum level RANK content low dose group,there was no statistically significant difference(P>0.05);Compared with OVX group,the serum RANKL content in the low and high dose groups at 4 weeks and 8 weeks of administration was reduced,with statistically significant differences(P<0.05).Compared with OVX group,serum E2 levels in the low and high dose groups at 4 weeks,8 weeks after administration were significantly increased(P<0.05).After 8 weeks of administration,the morphology of bone cells in the high and low dose group was observed by electron microscopy.Conclusion:1.Dendrobium officinalis enhanced the expression of estradiol,improved the bone metabolism of serum Ca and P,and achieved the effect of treating osteoporosis in ovariectomy rats.2.Dendrobium officinalis plays a role in the treatment of osteoporosis in ovariectomy rats by regulating the OPG/RANKL/RANK axis to inhibit osteoclast differentiation. |
