Objective: To study the effects of different concentrations of rosmarinic acid on human retinal capillary endothelial cells(HRCECs)proliferation and vascular endothelial growth factor(VEGF)expression in high glucose environment.Methods: Select HRCECs which fostered in vitro high sugar environment in good condition as the research object,the experimental were divided into low glucose control group(ML),high glucose control group(MG),high glucose +5umol/L rosmarinic acid group(M1),high glucose +25umol/L rosmarinic acid group(M2),high glucose +50umol/L rosmarinic acid group(M3),high glucose +100umol/L rosmarinic acid group(M4).The proliferation changes and the expression of VEGF of HRCECs were detected seperately by Methyl thiazolyl tetrazolium(MTT)and Western blot(WB)after the above concentration of rosmarinic acid acted on HRCECs for 24 hours,48 hours and 72 hours.Results :1.The MTT absorbance value of HRCECs cells after 24 hours,48 hours and 72 hours incubation in high glucose environment was significantly increased compared with that in low glucose environment(P<0.05).The longer the cells were cultured in vitro,the higher the light absorption value was measured by MTT.The gray value of VEGF in the high glucose environment was higher than that in the low glucose environment by WB(P<0.05),and the longer the incubation time was,the higher the gray value was.2.After 24 hours,48 hours and 72 hours of culture with different concentrations of rosmarinic acid added to HRCECs,the MTT absorbance value of cells in each treatment group was decreased compared with that in the high glucose control group,with statistical difference(P<0.05).At the same time,the higher the drug concentration was,the smaller the MTT absorbance value was.At the same concentration,the longer the action time,the smaller the absorption value.3.After HRCECs was cultured with rosmarinic acid at different concentrations for 24 hours,48 hours and 72 hours,compared with the high glucose control group,the gray value of VEGF in the cells of each treatment group decreased,with statistical difference(P<0.05).At the same time,the higher the drug concentration,the smaller the gray value.At the same concentration,the longer the action time,the smaller the gray value.Conclusion :1.Rosmarinic acid has an inhibitory effect on HRCECs cultured in high glucose environment,which is positively correlated with drug concentration and action time.2.Rosmarinic acid can reduce the expression of VEGF in HRCECs under high glucose environment,which is also positively correlated with drug concentration and action time.
