Objective:To explore the protective effect of β-asarone on H2O2-induced PC 12 cell and its underlying mechanism.Methods:MTT assay was used to detect the effect of different concentrations of H2O2 on the activity in PC 12 cells,and the appropriate concentration was used to construct the model of oxidative damage in PC 12 cells.MTT assay was used to detect the survival rate of PC12 cells pretreated by β-asarone.Accumulation of intracellular ROS was detected through DCFH-DA fluorescent probe,and the release of LDH and the activity of MDA,CAT,SOD and GSH of PC 12 cells pretreated by β-asarone were assessed.The protein and mRNA expression of Nrf2 and HO-1 were measured by RT-PCR and Western Blot.After transfected with a negative control siRNA and Nrf2 siRNA by small-interfering RNA kit to interfere with Nrf2 expression in PC 12 cells,MTT assay was used to detect the activity of PC 12 cells,and the activity of SOD and MDA in PC 12 cells pretreated by(3-asarone.Results:The treatment of β-asarone significantly alleviated the reduction in cell viability and the excessive accumulation of Lactate dehydrogenase(LDH),malondialdehyde(MDA)and reactive oxygen species(ROS)by increasing the activity of superoxide dismutase(SOD),catalase(CAT)and glutathione(GSH).Moreover,β-asarone pretreatment also activated nuclear factor 2 erythroid-related factor 2(Nrf2)and its downstream target heme oxygenase-1(HO-1),which was involved in quenching reactive oxygen to inhibit oxidative stress.Furthermore,when silenced by Nrf2 siRNA,the protective effect of β-asarone was reduced and the oxidative stress induced by H2O2 was enhanced.Conclusion:β-asarone could reduce oxidative stress via activating Nrf2/HO-1 pathway in PC 12 cells.
