| Objective:In this research,the quality standard of Momordica cochinchinensis was studied to establish a more perfect quality control system;At the same time,the commercial specification grades of Momordica cochinchinensis were studied.First of all,according to the actual market situation,under the guidance of the principle of "based on pharmacopoeia,based on the market,simple and practical",the commercial specifications and grades of the crude drugs were preliminarily divided,then,the correlation between the commercial specification grades and the system quantization index was analyzed in order to provide the theoretical basis for the high quality and high price in the market circulation process.Methods:1.Study on the quality standard of Momordica cochinchinensisBased on the provisions of Momordica cochinchinensis in the Chinese pharmacopoeia of 2015,routine examination items such as moisture,total ash and content determination items such as fatty oil and oleanolic acid were added.At the same time,HPLC fingerprints of the fruit and its decoction pieces were established.2.Preliminary classification of commercial specification grades of Momordica cochinchinensisThe length,width,thickness and single weight were selected as the quantitative indexes for the appearance traits of Momordica cochinchinensis.SPSS 19.0 statistical software was used to run K-mean clustering analysis on the measured results,Taking the central value of the final cluster as the reference value,combined with the non-quantifiable indexes such as color and degree of oil bleeding,and taking into account the actual operation,market demand and expert Suggestions,the commercial specification grades of Momordica cochinchinensis were preliminarily classified.3.Analysis on the correlation between the commercial specification grades and the systematic quantification index of Momordica cochinchinensisThe systematic quantitative index datas and fingerprints were collected and analyzed for different commercial specification grades of Momordica cochinchinensis,SPSS 19.0 statistical software was used to analyze the correlation between the appearance character,commercial specification grades and internal index of Momordica cochinchinensis.Results:1.(1)The results of morphological identification,microscopic identification,TLC identification and content of Gypsogenin 3-O-β-D-glucuronic Acid Methyl Ester all comply with the requirements of pharmacopoeia.(2)The water content of the seeds of Momordica cochinchinensis shouldn’t exceed 6.0%,and the total ash content shouldn’t exceed 3.0%.The water content of defatted Momordica cochinchinensis seed powder shouldn’t exceed8.0%,and the total ash content shouldn’t exceed 6.0%.(3)The fingerprints of Momordica cochinchinensis and the defatted Momordica cochinchinensis powder were established,14 common peaks were matched and oleanolic peak(no.11 peak)was selected as the reference peak.The similarity of fingerprint between different batches of Momordica cochinchinensis was greater than 0.950,and the similarity of fingerprint between different batches of the defatted Momordica cochinchinensis powder was greater than 0.950.(4)The content of fatty oil and oleanolic acid of Momordica cochinchinensis should not be less than 35.0% and 0.015% respectively.The fatty oil content range of the defatted Momordica cochinchinensis powder is 16% ~ 24%,and the oleanolic acid content should not be less than 0.022%.2.The color,oil bleeding degree and single weight of kernel were used as the distinguishing points,Full seed,white seed,no oil situation as the selection of goods,according to the quantitative index,the weight of the first grade is greater than 4 g,and the weight of the second grade is between 3 and 4g.There is no regulation on the traits of the uniform Ly-priced goods.3.(1)The results of internal quality determination of Momordica cochinchinensis of different commercial specifications and grades showed that all the samples met the quality standards of Momordica cochinchinensis.Moisture: first grade < second grade < unified goods;Total ash content:second grade < first grade< unified goods;Content of fatty oil : unified goods< second grade < first grade;Content of Gypsogenin 3-O-β-D-glucuronic Acid Methyl Ester: unified goods < first grade = second grade;oleanolic acid :unified goods = second grade < first grade,However,the water content,total ash content,fatty oil,Gypsogenin 3-O-β-D-glucuronic Acid Methyl Ester and oleanolic acid were overlapped.(2)The HPLC fingerprints of the samples with different commercial specifications and grades showed high similarity.(3)There was a significant positive correlation between width and fatty oil content,and a significant positive correlation between single weight and oleanolic acid content.Momordica cochinchinensis commercial specifications and grades and inner index correlation analysis showed that fatty oil content and grades were significant positive correlation,other indexes had no significant correlation.The results of multiple comparative analysis showed that the selection of fatty oil content was larger than that of general goods,and there was a significant difference in the fatty oil content between first grade goods and unified goods,There was no significant difference in the fatty oil content between the first and second grades of the selected goods,and no significant difference in the other indexes.Conclusion:Through the increase of the inspection items for the quality standard of Momordica cochinchinensis,the improvement of the quality standard system for it was realized,and the quality standard for Momordica cochinchinensis was drafted.This standard can accurately and effectively control the quality Momordica cochinchinensis.The classification standard of the commercial specifications and grades of Momordica cochinchinensis was preliminarily established.The results of the research on the correlation between the commercial specification grades and the internal quality index of Momordica cochinchinensis provide a theoretical basis for the commercial specification and grade standard which was stablished on the basis of "Quality assessment based on feature identification". |
