ObjectiveLncRNAs with abnormal expression in Peripheral Blood Mononuclear Cells(PBMC)of the patiens with esophageal squamous cell carcinoma were screened and verified,and the relationship between the expression level and the clinicopathological characteristics of ESCC patients was comprehensively analyzed in combination with the clinical data of patients,so as to provide potential non-invasive biomolecular detection indicators for the diagnosis of ESCC and provide new candidate targets for the treatment.Methods1.Five cases of normal physical examination patients and five cases of esophageal squamous cell carcinoma patients who were pathologically diagnosed after surgery and had not received radiotherapy or chemotherapy were selected.Peripheral blood mononuclear cells were extracted for RNA sequencing.2.In the later stage,40 esophageal squamous cell carcinoma patients without radiotherapy and chemotherapy and 30 normal subjects were selected to verify the differential expression of SNHG3 and SNHG20 in PBMC of esophageal squamous cell carcinoma patients and normal subjects by qRT-PCR.3.The clinical and pathological characteristics of patients were obtained by the hospital HIS system.To find the rule,the relationship between SNHG3 and SNHG20 relative expression levels was statistically analyzed by SPSS and GraphPad statistical software.Result1.Compared with PBMC of normal subjects,158 differential expression lncRNAs(differential expression lncRNA,DEL)were identified(54 upregulate and 104 downregulate)in the PBMC of patients with esophageal squamous cell carcinoma in this study,with P<0.05.158 differentially expressed lncRNAs are widely involved in a variety of diseases and signalling pathways.Among all DELs,the most significantly down-regulated lncRNAs in PBMC of patients with esophageal squamous cell carcinoma were SNHG20 and SNHG3,ranking first and second,P<0.001.2.The relative expression of SNHG3 in PBMC of 40 patients with esophageal squamous cell carcinoma was 0.587±0.870,and that in normal subjects was 2.520± 2.112,the former was significantly lower than the latter,and the difference was statistically significant(t=4.723,P<0.001).At the same time,the relative expression of SNHG20 in PBMC of ESCC patients was 0.907±1.688,significantly lower than 2.550±2.590 in normal subjects,with statistically significant difference(t=3.025,P<0.001).3.Statistical analysis was performed on the relationship between the changes in the relative expression levels of SNHG3 and SNHG20 and the clinicopathological factors in ESCC patients.It was found that SNHG3 and SNHG20 in PBMC had different effects on ESCC.The down-regulated expression of SNHG3 in PBMC was closely related to tumour size,lymph node infiltration,whether there was distant metastasis and TNM stage.What’s more,the difference between the two groups was statistically significant(P<0.05).But,between patients of different ages,genders and tumour differentiation degrees,there was no statistically significant difference in the expression of SNHG3 in PBMC.The decreased expression level of SNHG20 in PBMC was closely related to the differentiation degree of ESCC,lymph node infiltration,whether there was distant metastasis and TNM stage.Furthermore,the difference between the two groups was statistically significant(P<0.05).On the contrast,there was no significant difference in sex,age or tumour size.ConclusionThe expression levels of SNHG3 and SNHG20 in PBMC of patients with esophageal squamous cell carcinoma were significantly lower than those in normal subjects,and further analysis showed that the abnormal expression levels of SNHG3 and SNHG20 in PBMC of ESCC patients were closely related to some clinicopathological characteristics of esophageal squamous cell carcinoma,respectively.The results of this study suggest that SNHG3 and SNHG20 is expected to become new non-invasive detection markers esophageal squamous carcinoma and potential therapeutic targets.
