Study On Chemical Constituents And Sleeping Pharmacological Activity Of Gastrodia Elata Bl. Effective Parts

Gastrodia elata is a dried tuber of Orchidaceae Gastrodia elata Bl.It is sweet and flat,has the effects of relieving wind and stopping spasm,calming liver yang,and eliminating wind and collaterals.It is a traditional and precious Chinese herbal medicine.Modern research shows that phenolic acids and polysaccharides are the main active ingredients of Gastrodia elata,and they have very good effects on the central nervous system and circulatory system.The effect of sedation and hypnosis is more significant.Therefore,this research group takes Gastrodia elata as the research object,and conducts in-depth research on its pharmacological substance basis for improving sleep.Objective: In this project,the autonomic activity model of mice and the synergistic experimental model of sodium pentobarbital were used to screen different solvent extracts and different polar extracts of Gastrodia elata to obtain effective parts of Gastrodia improving sleep;and to study the mechanism of effective parts;The two effective parts of Gastrodia better sleep are the research objects.The preliminary exploration of medicinal substances using modern separation and identification and spectral analysis methods provides a solid theoretical basis for the further development and utilization of Gastrodia medicinal resources.Methods: 1.Taking Gastrodia elata as the research object,through the mouse’s autonomous activity,the mice were induced to sleep on experimental models of sodium pentobarbital above and below the threshold dose,and different solvent extracts of Gastrodia were screened;the same pharmacological experimental model was used to screen Gastrodia alcohol extracts Extracts with different polarities to get effective parts of Gastrodia sleep improvement.2.ELISA was used to determine the content of Glu,GABA,5-HT and DA in the hypothalamus and hippocampus of rats.The expression of GABAARα1,GABAARγ2-positive cells in brain tissue was observed by immunohistochemistry,and the role of Gastrodia in improving the effective part of sleep Mechanism;3.Using silica gel column chromatography,Sephadex LH-20 column chromatography and semi-preparativehigh performance liquid chromatography and other modern extraction and separation technologies to systematically study the effective sleep improvement parts of Gastrodia elata,and comprehensively use IR,MS,NMR And other modern spectroscopy methods and techniques for structural identification of the isolated active ingredients.Results: 1.According to the efficacy screening,the Gastrodia ethanol extract has good sleep improving activity.It was extracted with different polar solvents,and the results showed that ethyl acetate extract had the best effect on improving sleep activity,followed by n-butanol extract.2.In the study of improving sleep mechanism,it was found that ethyl acetate extract can significantly increase the levels of GABA,5-HT,DA in the hypothalamus and hippocampus of insomnia model rats,reduce the level of Glu in the hypothalamus,protect the hypothalamus,Hippocampal neuronal cells enhance the expression of GABAAR α 1 and GABAAR γ 2-positive cells in the hypothalamus and hippocampus.3.Using modern Chinese medicine extraction and separation technology and spectral identification technology to isolate and identify 18 compounds from Gastrodia improving sleep activity ethyl acetate and n-butanol extract,which are stigmasterol,syringic acid,vanillin,carotene,p-hydroxyl Benzaldehyde,vanillyl alcohol,p-hydroxybenzyl alcohol,vanillic acid,m-hydroxybenzoic acid,p-hydroxybenzoic acid,gastrodin,parisonsin B,adenosine,parisons C,caffeic acid,protocatechuic acid succinic acid,palmitic acid,of which caffeic acid was isolated from Gastrodia elata for the first time,which provided a reliable theoretical basis for further basic research of medicinal substances.Conclusion: Gastrodia ethyl acetate extract has a better sleep improvement effect,and its mechanism may be related to regulating the content of monoamine neurotransmitters in the hypothalamus and hippocampus of rats;the effective parts of improving sleep are separated and identified,and most of the compounds It is a phenolic acid and its glycoside compounds,and it has been verified that the active ingredients of Gastrodia elata improving sleep are mainly phenolic compounds.
