| Objective: To observe the effect of LNK on vascular smooth muscle cells(VSMC)and inflammatory cytokines in hypertension,and to explore the role of LNK in vascular remodeling in hypertension.Methods: Rat thoracic aorta smooth muscle cells were selected to construct a model of hypertensive cells under certain conditions.According to different treatment methods,it was divided into control group,blank group,no-load group and LNK group.The levels of VSMC cycle,apoptosis and proliferation were detected by flow cytometry.ELISA was used to detect the expression levels of IL-6,TNF-α and IFN-γ.Results:(1)levels of vascular smooth muscle cell cycle,apoptosis and proliferation in each group: compared with the control group,VSMC in the blank group and the no-load group decreased in G0/G1 phase,increased in S phase,and increased in G2/M phase,with statistically significant differences(P<0.05)Compared with the no-load group,VSMC in LNK group increased in G0/G1 phase,decreased in S phase,and decreased in G2/M phase,with statistically significant differences(P<0.05).The apoptosis rate of VSMC cells in LNK group was higher than that in the blank group and the no-load group(P<0.05).The proliferation level of VSMC cells in the blank group and the no-load group was higher than that in the control group(P<0.05).The proliferation level of VSMC cells in LNK group was lower than that in the blank group and the no-load group(P<0.05).(2)the levels of inflammatory cytokines on vascular smooth muscle cells in each group: compared with the control group,the levels of IL-6,TNF-α and IFN-γ were increased in the blank group and the no-load group,and the expressions of IL-6 and IFNwere decreased in the LNK group,with statistically significant differences(P<0.05).Compared with the control group,the expression of IL-6,TNF-α and IFN-γ in the no-load group was increased,and the difference was statistically significant(P<0.05).Compared with the blank group and the no-load group,IL-6,TNF-α and IFN-γexpression were decreased in the LNK group,and the difference was statistically significant(P<0.05).Conclusion: LNK is involved in vascular remodeling in hypertension by mediating inflammatory response to affect vascular smooth muscle cycle,apoptosis and proliferation. |
