Objective: Screening early diagnosis biomarker proteins of acute aortic dissection by4 D Label-free quantitative proteomics technology,and realizing early and rapid diagnosis of acute aortic dissection.Methods: This experiment used Case-control study method,selecting 10 patients with Stanford type A acute aortic dissection and 10 patients with coronary heart disease who underwent coronary artery bypass graft.Ascending aortic tissue specimens was taken during the operation.Firstly the changes of proteomic profiles of active tissue samples from two groups of patients were analysed,complete protein identification and quantification,screening for significantly differentially expressed proteins were performed by 4D Label-free quantitative proteomics technology.Further database search was performed to analyze the biological information of the differentially expressed proteins.Based on the results of bioinformatics analysis,seven significant differentially expressed proteins related to the occurrence and development of acute aortic dissection were selected.Finally,the receiver operating characteristic curve method was used to analyze the diagnostic value of the seven differential proteins for acute aortic dissection.Results:1.A total of 3985 proteins were identified,of which 3350 proteins were quantifiable.2.4D Label-free technology were used to calculate the differential expression of proteins between the two groups,there were 139 significantly highly expressed proteins with the fold change≥1.5 times,and 108 significantly low-expressed proteins with the old change ≤ 0.67 times.3.Results of Bioinformatics analysis of significant differential proteins show that the functions of differential proteins are mainly related to biological processes such as inflammatory response,apoptosis,degradation of extracellular matrix,cell metabolism,stress response,and TGF-β signaling pathway.4.The comparison of the diagnostic value of 7 differential proteins for AAD shows that,except for Decorin,the AUC of the other 6 differential proteins are all>0.70,all of whichhave high sensitivity and specificity,and may be used as early diagnostic markers of acute aortic dissection.Conclusions: In this study,we used high-throughput 4D Label-free quantitative proteomics technologies to screen for significant differential proteins,finally it has been preliminarily identified that Six proteins including LTBP1,TGFB1,SAA1 may be used as early candidate diagnostic biomarkers of acute aortic dissection.
