Objective:This study expects to test tissue differences and change rules of the expression level of SATB1 and DNMT1 protein in different ovarian tissue,both in normal ovarian tissue,benign ovarian epithelial tumors and malignant ovarian epithelial tumors by immunohi-stochemical staining SP method,analyse if SATB1 protein and DNMT1 protein expression level in various organizations have relationship,further to find the mechanism on ovarian tissue of the SATB1 protein and DNMT1 protein may play in the process of malignant transformation.Methods:Using chi-square test and rank correlation analysis their different expression in epithelial ovarian malignant tumor tissues,and make statistical research for the relationship between them and the biological behavior of ovarian cancer.Results:1.The positive expression of SATB1 protein in 10 cases of normal ovarian tissue,30cases of ovarian benign epithelial tumors and 50 cases of malignant tumor tissues were in 1case,10 cases,35 cases,the positive expression rate was 10%,33.3%and 70.0%in turn.The positive expression rate of ovarian malignant tumor group compared with the other two groups,the difference is statistically significant(x~2=16.06,P<0.05),and ovarian benign epithelial tumor group compared with normal group,there was no statistically significant difference(x~2=2.05,P>0.05).2.The positive expression of SATB1 protein in ovarian cancer tissue was associated with TNM staging,SATB1 protein expression rate in early stage and late stage were 45.5%and 89.3%respectively,their comparative difference was statistically significant(x~2=11.27,P<0.05).3.SATB1 protein expression levels was positive correlated with pathological classification,the significant defferences of SATB1 protein expression in ovarian cancer tissue samples exist between low grade group and high grade group.Low grade group considerably below high grade groups(x~2=12.96,P<0.05).4.There is no significant difference of the expression rate of SATB1 protein in different age groups of patients with ovarian cancer specimens,no statistical significance(x~2=3.70,P>0.05);5.Among the clinical specimens of patients with different pathological types of ovarian cancer,there was no statistically significant difference of SATB1 protein expression rate in each group(x~2=1.79,P>0.05).6.There was no statistically significant difference of SATB1 protein expression rate between no lymph node metastasis and lymph node metastasis group of patients specimen,which were confirmed by postoperative pathology(x~2=0.61,P>0.05).7.There was no statistically significant difference of the positive expression rate of SATB1 protein between ascites and no ascites group(x~2=0.52,P>0.05).8.The positive expression of DNMT1 protein in 10 cases of normal ovarian tissue,30cases of ovarian benign epithelial tumors and 50 cases of malignant tumor tissues were in 1case,9 cases,30 cases,the positive expression rate was 10%,30%and 60.0%in turn.The positive expression rate of ovarian malignant tumor group compared with the other two groups,the difference is statistically significant(x~2=11.03,P<0.05),and ovarian benign epithelial tumor group compared with normal group,there was no statistically significant difference(x~2=1.60,P>0.05).9.The positive expression of DNMT1 protein in ovarian cancer tissue was associated with TNM staging,DNMT1 protein expression rate in early stage and late stage were36.4%and 78.6%respectively,their comparative difference was statistically significant(x~2=9.15,P<0.05).10.DNMT1 protein expression levels was negative correlated with pathological classification,the significant defferences of DNMT1 protein expression in ovarian cancer tissue samples exist between low grade group and high grade group.Low grade group considerably below high grade groups(x~2=8.33,P<0.05).11.There is no significant difference of the expression rate of DNMT1 protein in different age groups of patients with ovarian cancer specimens,no statistical significance(x~2=2.38,P>0.05);12.Among the clinical specimens of patients with different pathological types of ovarian cancer,there was no statistically significant difference of DNMT1 protein expression rate in each group(x~2=1.44,P>0.05).13.There was no statistically significant difference of DNMT1 protein expression rate between no lymph node metastasis and lymph node metastasis group of patients specimen,which were confirmed by postoperative pathology(x~2=2.20,P>0.05).14.There was no statistically significant difference of the positive expression rate of DNMT1 protein between ascites and no ascites group(x~2=0.05,P>0.05).15.There was a moderate linear positive correlation between the expression of SATB1protein and DNMT1 protein(r=0.407,x~2=9.92 P=0.002).Conclusion:1.High expression of SATB1 and DNMT1 is closely related to the occurrence of ovarian cancer,and they participated in the early process of ovarian cancer.The expression rate is gradually increasing n normal ovarian tissue,benign ovarian tumor and in ovarian cancer,it is expected to be as one of the important molecular marker of predicting ovarian malignant change.2.The rise of SATB1 and DNMT1 expression is associated with pathological grading and clinical staging of ovarian cancer progression,may be closely with ovarian cancer invasion and infiltration,can be used as evaluation of the degree of the clinical course of patients with ovarian cancer,it is expected to be the biological target predicting potential infiltration trend.3.There is no significant correlation between the age of the patients with ovarian cancer and the occurrence of diseases,the distribution characteristics of SATB1 and DNMT1 in different age groups were found no statistically significant.4.The expression of SATB1 and DNMT1 proteins is irrelevant to the lymph node metastasis and the ascites cancer cells have no specificity,the positive expression rate of each group were at higher levels,it prompts abnormal secretion of SATB1 and of DNMT1can promote the growth of cancer cells,they infringe other organizations before lymph node metastasis,maybe associated with other transfer way that including direct spreading and celiac planting.5.In the process of ovarian tumor occurence,coordination with DNMT1 of SATB1expression play an important role in promoting,combined detection can improve the positive detection rate of ovarian cancer. |
