Association Of Human Leukocyte Antigen Ⅱ Gene Polymorphism With Graves’ Disease And Its Interaction With Helicobacter Pylori Infection

Objectives:To investigate the correlation between human leukocyte antigen II gene polymorphism and Graves’ disease(GD),the relationship between Helicobacter Pylori(HP)infection and GD,and the effect of the interaction of HLA-II genes with HP infection on the pathogenesis of GD.Methods:A total of 288 patients diagnosed with GD were recruited,112 healthy volunteers constituted the control group.Molecμlar typing of the 50 locis in HLA-Ⅱgene were conducted using polymerase chain reaction-sequence based typing method and polymerase chain reaction with sequence specific primers method.Levels of HP immunoglobulin antibodies were detected using enzyme-linked immunosorbent assaysResults:The frequency of HLA-DPB1*0501 gene in GD group was significantly higher than that in control group,which was statistically different between the two groups(P=0.015);The frequencies of HLA-DRB 1*0701,HLA-DRB 1*1202,HLA-DRB1*1302,HLA-DQB1*0501 and HLA-DQB1*0503 genes in GD group were significantly lower than that in control group,which were also statistically different between the two groups(P=0.010,P=0.000,P=0.006,P=0.000,P=0.004).The HP infection rate of GD group was significantly higher than that in control group,and the two were statistically different(P=0.000);The odds ratio of GD patients infected with HP,and positive HLA-DPB 1*0501 or negative HLA-DRB 1*0701 or negative HL A-DRB1*1202 or negative HLA-DRB 1*1302 or negative HLA-DQB 1*0501 or negative HLA-DQB 1*0503 were significantly higher than those with only one of these indices.Conclusions:HLA-DPB 1*0501 may be a susceptibility gene in GD patients;HLA-DRB1*07015 HLA-DRB 1*1202,HLA-DRB1*1302,HLA-DQB 1*0501,HLA-DQB 1*0503 may be protective genes for GD patients.HP infection is associated with GD.The different combinations of HP infection with HLA-Ⅱ alleles had different effects on the risk of GD.The combination of HP infection and positive HLA-DPB1*0501 or negative HLA-DRB 1*0701 or negative HLA-DRB 1*1202 or negative HLA-DRB 1*1302 or negative HLA-DQB 1*0501 or negative HLA-DQB 1*0503 genotypes may increase the risk of GD.
