Objective:The study is to investigate the effect of circELP3 on the proliferation of bladder cancer cell lines,and to verify it in vivo in animals.To verify the expression of circELP3 in clinical tissue samples,and to analyze the relationship between circELP3 expression and clinical sample with its stage and grade.To provide a new marker for diagnosis of bladder cancer in future,and to provide a new method and an idea for treatment of bladder cancer in the future.Method:1.Transfection of siRNA sequences into bladder cancer cells by cell transfection experiments in vitro,silence the expression level of circELP3,and verify the efficiency of siRNA sequences2.By reducing the expression level of circELP3,cck8 assay was used to detect its effect on the proliferation of bladder cancer cell lines,and the colony formation assay was used to evaluate its effect on the colony forming ability.3.Verify the localization of circELP3 in bladder cancer cell lines by fluorescence in situ hybridization.4.Based on the siRNA sequence,sh-circELP3 was synthesized and the sh-circELP3 T24 cell line was constructed,then we performing a subcutaneous tumor-forming animal experiment,to verify the effect of silence circELP3 on the proliferation of bladder cancer in vivo.5.Through real-time quantitative PCR experiment,detecting the expression levels of circELP3 in 23 pairs of bladder cancer specimens and adjacent specimens,and analyzing the expression levels of circELP3 between the cancer specimens and adjacent specimens.6.Through real-time quantitative PCR experiment,detecting the expression levels of circELP3 in 30 clinical specimens of bladder cancer,analyzing the relationship between the expression levels of circELP3 and clinical stage or grade.Result:1.In vitro,the expression level of circELP3 was significantly reduced by transfection of siRNA sequence,and the difference was statistically significant(P<0.05).2.In vitro,transfection of siRNA sequence,can significantly inhibited the proliferation of bladder cancer T24 cell line and 5637 cell line.The difference was statistically significant(P<0.01).3.In vitro,transfection of siRNA sequence,can significantly inhibited the colony forming ability of bladder cancer T24 cell line and 5637 cell line,and the difference was statistically significant(P<0.01).4.In bladder cancer T24 cell line and 5637 cell line,circELP3 is mainly expressed in the cytoplasm of cells.5.Animal experiments showed that sh-circELP3 group can significantly reduced subcutaneous tumor volume and weight compared with the sh-nc group,the difference was statistically significant(P<0.05).6.In 23 pairs of clinical specimens,the expression levels of circELP3 in bladder cancer specimens was higher than that adjacent specimens,and the difference was statistically significant(P<0.05).7.In the TMN stage of 30 clinical bladder cancer specimens,the expression levels of circELP3 in T3-4 patients was higher than that in Ta,T1 and T2 patients,and the difference was statistically significant(P<0.05).The expression levels of circELP3 in N1-3 clinical bladder cancer specimens was higher than that in NO clinical bladder cancer specimens,and the difference was statistically significant(P<0.05).There was no significant correlation between the expression levels of circELP3 and the grade of tumor in clinical bladder cancer specimens,P>0.05.Conclusion:In vitro,silencing the expression levels of circELP3 can significantly inhibit the proliferation and colony forming ability of bladder cancer T24 cell line and 5637 cell line.The expression of circELP3 levels in bladder cancer tissues is higher than that in bladder cancer adjacent tissues,The T3-T4 expression levels of circELP3 in patients was higher than that in patients with Ta,T1 and T2.The N1-3 expression levels of circELP3 in patients was higher than that in patients with N0,these res μ It suggesting that circELP3 may act as a tumor-promoting gene in bladder cancer,and it maybe playing an important role in cancer progress of bladder cancer.so It maybe can become a new biomarker for the diagnosis of bladder cancer,and maybe become a new method for the treatment of bladder cancer.
