In recent years,the rapid development of plasma biomedicine has formed two research routes:direct plasma treatment and indirect plasma treatment.Plasma direct treatment mainly relies on various therapeutic devices based on plasma discharge to interact with organisms directly.The indirect product,such as plasma activated water,acts primarily with the organism.However,the life of plasma-activated water is extremely limited.Whether plasma can be used to generate a long-life,low-cost,easy-to-use,and environmentally friendly plasma activator is a key issue to be solved based on the research direction of plasma indirect therapy.The olive oil was used as the raw material to be treatd by plasma source to obtain the plasma activated oil.The basic properties and composition of the plasma activated oil were quantitatively and qualitatively analyzed by chemical reagent detection and physical spectroscopy.Based on the research of plasma activated oil sterilization,the mouse skin wound healing experiment was carried out.The expression of cyclin(CD34)and vascular endothelial growth factor(VEGF)affected by plasma-activated oil was further measured with immunohistochemical analysis.Finally,the mechanism of activated oil on promoting skin wound healing in mice was summarized.(1)Plasma active oil is a third-generation oil-based drug and the preparation method is simple.The plasma jet process which is low-cost and environmentally friendly is used to treat cheap olive oil under atmospheric pressure and room temperature conditions.The plasma jet surface discharge produces many high energy particles and reactive oxygen atoms at the plasma and oil surface interface.The dissociation of C=C double bonds and the oxidation of reactive oxygen atoms in fatty acids by those high energy particles are the main mechanism for the production of H2O2 active particles and carboxylic acids in plasma-activated oils.(2)By measuring the physical and chemical properties of the plasma-activated oil,the peroxide value and acid value of the plasma-activated oil are 7.5 times and 57%,respectively,of the conventional ozone oil.At the same time,the plasma-activated oil exhibits excellent preservation properties.Unlike plasma-activated water with a shelf life less than one week,the plasma-activated oil can be stored at room temperature for at least3 months,the shelf life of plasma-activated oil is expected up to 1 year.(3)Plasma-activated oil can not only kill bacteria on the wound,but the carboxylic acid in the plasma-activated oil can cause cell membrane rupture,inhibit the necessary metabolic reaction,and imbalance the intracellular pH.The peroxide can produce a large amount of oxygenated particles with biological activity and cytotoxicity,the strong oxidative properties can cause oxidation and inactivation of biomolecule.Studies have shown that carboxylic acids and peroxides can also promote the expression of growth factors such as VEGF and CD34.Therefore,the healing rate of wounds treated with plasma-activated oil is 28.5%faster than that of the control group.Plasma activated oils are suitable for a variety of biomedical applications,such as chronic wounds of diabetic wounds and pathology of vaginal mucosa,while plasma activated oil production methods are scalable and suitable for future medium scale industrial production. |
