Objective To determine whether miR-200 c can reserve the process of epithelial mesenchymal transformation(EMT)in gastric carcinoma cells and search the correlation between the reservation of miR-200 c on the process of EMT and PI3K/AKT signaling pathway in gastric carcinoma cells.Methods Gastric cancer cell lines with moderate expression of miR-200 c were screened and corresponding drug-resistant gastric cancer cells were selected as experimental target cells by detecting the expression level of miR-200 c in different gastric cancer cell lines.The differences of cell morphology,migration and invasion ability,EMT marker protein,miR-200 c and PI3K/AKT signaling pathway related genes and proteins were compared between the two groups.The drug-resistant gastric cancer cells were used as the target cells and artificially overexpressed miR-200 c by lentivirus transfection.After the successful transfection,the changes of cell morphology,migration and invasion ability,EMT marker protein and PI3K/AKT signaling pathway related genes and proteins were compared.Results (1)Based on the expression level of BGC-823,the expression levels of miR-200 c in seven gastric cancer cells lines were BGC-823=1,MKN-45=1.069,HGC-27=1.831,SGC-7901=8.242,AGS=300.871,MGC-803=342.509,MKN-74=1181.169,and the differences were statistically significant(P<0.01).(2)In the drug-resistant cells,SGC-7901/ADR,the ability of invasion migration and resistance to adriamycin were enhanced,and the expression of EMT-related protein E-cadherin was decreased,but the expression of Vimentin was increased;the expression of miR-200 c was decreased while the expression of PI3K/AKT signal pathway related genes PI3 K,AKT,mTOR,P70S6 K and 4-EBP were increased,and the expression levels of p-AKT and p-P70S6 K related activating proteins increased.The difference were all statistically significant(P < 0.05).(3)In group of SGC-7901/ADR-V,transfected with miR-200 c,the susceptibility of cells to adriamycin was significant stronger than the group of SGC-7901/ADR.At the same time,the invasion and migration ability of cells was significantly weakened.The expression of E-cadherin was significantly increased while the Vimentin was significantly decreased.The expression levels of PI3 K,AKT,mTOR,P70S6 K,4-EBP,p-AKT and p-P70S6 K,related genes and proteins in PI3K/AKT signaling pathway,were also significant decreased(P<0.01).Conclusions Overexpressed miR-200 c may play an indirect role in inhibiting the EMT process in gastric cancer cells through PI3K/AKT signaling pathway. |
