Objective:In this paper,the seeds of Psoralea corylifolia L.were extracted and separated with biological activity as the guide,and the effective active ingredients were screened.Then the chemical structures of the compounds were determined according to the relevant spectral and magnetic resonance data,and the effective inhibitory concentrations of protein tyrosine phosphatase 1B(PTP1B)and diethylglycerol acyltransferase(DGAT)were determined.Methods:Through reading the relevant literature,refer to its experimental methods.The effective chemical constituents in Psoralea corylifolia L.seeds were separated and purified by normal-phase silica gel column chromatography,C188 reversed-phase columnchromatographyandSemi-preparativehighperformanceliquid chromatography.The chemical structure of the compounds was determined by analyzing the spectral data of 1H-NMR,13C-NMR and HMBC,and comparing with the spectral data in relevant literature.Results:The chemical constituents of 75%ethanol extract from Psoralea corylifolia L.seeds were extracted and separated.Nine flavonoids were obtained,including two new compounds and seven known compounds.Based on the analysis of NMR data and references,the structures of compounds 1-9 were determined as follows:3"-methoxy-bavacoumestan C(1),6,7-furanbavachinone B(2),bavachinone B(3),bavachin(4),bavachinin(5),bavachinone A(6),brosimacutin E(7),neo-bavaisoenaavone(8),corylifol A(9).Conclusion:Compounds 1-9 were isolated from Psoralea corylifolia L.seeds.Compounds 1-2 were new compounds and compounds 3-9 were known compounds.Compounds 1,4,5,8 and 9 selectively inhibited PTP1B and DGAT.The effective inhibitory concentration(IC50)ranged from 9.1±1.1μM to 21.3±1.1μM and from78.5±1.7μM to 153.4±1.5μM,respectively. |
