Objective: The study is to determine and compare the serum levels of GADA,ICA,IAA,IA2 A and ZnT-8A in type 1 diabetic patients and healthy people by western blotting and chemiluminescence.To explore the diagnostic performance of the five antibodies in these two methods and to evaluate which method is more efficient and more suitable for clinical type 1 diabetes diagnosis.Methods: The subjects were divided into two groups.one group was type 1 diabetes group,the other group was healthy control group,150 cases in type 1 diabetes group and 120 cases in healthy control group.The levels of GADA,ICA,IAA,IA2 A and ZnT-8A in the two groups were detected by western blotting and chemiluminescence respectively.The detection rates of these markers by western blotting and chemiluminescence were analyzed by chi-square test and ROC curves.Results: The positive rates of IAA,GADA,IA2 A,ICA and ZnT-8A in the type 1 diabetes group by chemiluminescence were 15.3%,56.7%,36%,41.3% and 26.7%,the positive rates of IAA,GADA,IA2 A,ICA and ZnT-8A in the type 1 diabetes group by western blotting were 2.7%,12.7%,18.7%,0.7% and 15.3%,the difference was statistically significant in this two mothed,P<0.05.The chi-square test and ROC curve analysis both showed that the chemiluminescence method was more sensitive than the western blotting for detecting 5 antibodies in diabetes,and the positive rate was higher when the combined detection of antibodies.Conclusion: Detection islet autoantibodies is helpful for the diagnosis of type 1 diabetes.Chemiluminescence detection sensitivity of diabetic autoantibodies compared to western blotting high more conducive to the diagnosis of type 1 diabetes and combined detection can increase the positive detection rate.
