EGCG is the main active substance of tea and has strong antioxidation and anti-cancer effects.However,its bioavailability in the human body is low.Phyllanthus emblica is a medicinal and edible tropical fruit.Emblica polysaccharide is one of the active ingredients of its biological activity,and it also has anti-oxidation and anti-cancer effects.In this paper,Phyllanthus emblica and its extracts were used as the carriers to explore the isothermal adsorption and adsorption kinetics characteristics,and the enhancement on bioavailability of the carriers in vitro simulated digestion experiments.Explore the anti-cancer effect of combination of EGCG and emblica polysaccharide.The main contents are as follows: 1.Study on Adsorption of EGCG by Phyllanthus emblica Fruit PowderThe adsorption kinetics(pesudo-first-order,pesudo-second-order kinetic equation,Weber-Morris particle diffusion model),adsorption thermodynamics,isothermal adsorption models(Langmuir & Freundlich)were employed to describe the the adsorption behaviors of EGCG onto Phyllanthus emblica fruit powder.The results showed that pseudo-secondorder was fitted better to experimental data.At 25 ℃,it has uniform adsorption and can reach the theoretical maximum adsorption capacity of 17.15 mg / g.In thermodynamic analysis,ΔH° was-18.99 kJ/mol and ΔS° was 1.37 J/mol/K,indicating that the adsorption process was endothermic,and ΔG° was between-19.33 and-19.59 kJ/mol with ambient temperature ranged from 25 to 55 ℃,exhibiting that the adsorption process was predominantly manipulated by a physisorption mechanism.2.The adsorption of EGCG onto pretreatment Phyllanthus emblica fruit powder and In vitro simulated digestion experiment.The three types of pretreatment of Phyllanthus emblica fruit,namely citric-sodium bicarbonate(CSP),ethanol(EP)and NaOH-HCl(NHP)pretreated powder,have theoretical maximum adsorption capacity at 25 ° C,which were 56.81,30.96,51.28 mg/g 3,respectively.According to thermodynamic analysis,the adsorption is a spontaneous and exothermic process dominated by physisorption adsorption.The results of in vitro simulated digestion showed that the effect of CSP was most efficient carrier for EGCG delivery,with coverery rate of 52% of the intitial EGCG amounts.3.Anticancer effect of EGCG combined with Emblica polysaccharideHepG-2 cells were used as experimental subjects for anticancer experiments.MTT proliferation assay showed that EGCG combined with Emblica polysaccharide could inhibit the proliferation of HepG-2 cells.Under the treatment of EGCG and Emblica polysaccharide were 2000 μmol/L and 500 μg/mL,respectively,for 48 h,the inhibition rate was 29.68%;DCFH-DA experiments showed that EGCG combined with Emblica polysaccharide,which could increase the intracellular reactive oxygen species and damage the cells.The results of mitochondrial membrane potential measurement indicated that EGCG was combined with Emblica polysaccharide;The amount of red fluorescent expression in the mitochondria of the treated liver cancer cells was greatly reduced,and the mitochondria of the hepatoma cells were damaged.The results of RNAseq analysis showed that HepG-2 cells treated with EGCG and Emblica polysaccharide and untreated HepG-2 cells Compared with,there were 9081 genes with significant differences,of which 2163 significantly upregulated the genes and 6918 significantly down-regulated the genes. |
