| Objective: Colorectal cancer(CRC)is the third most commonly diagnosed malignant tumor in the world,with the morbidity and mortality increasing over the recent years in China.Myosin Ⅵ(MYO6)is involved in many physiological activities as cell dynein.Previous studies suggested that myosin Ⅵ is highly expressed in cervical cancer,prostate cancer and other malignant tumors,and is involved in the regulation of tumor cell growth and apoptosis.This study aimed to analysis the impacts of myosin VI on the growth and apoptosis of colorectal cancer cells.Methods: 1.We analyzed the expression level of myosin Ⅵ in colorectal cancer cells in Oncomine database.2.We established stably MYO6 knockdown SW1116 colon cancer cells via lentivirus infection.Then by polymerase chain reaction(Polymerase,Chain Reaction,PCR)and Western blot(Western blot WB),we detected the expression level of MYO6 in lentivirus infected SW1116 cell line.3.The impacts of Myosin Ⅵ on the growth and apoptosis of SW1116 cells were tested through MTT assay,colony formation assay,cell cycle analysis and apoptosis analysis.Results: 1.MYO6 was highly expressed in colorectal cancer specimen.2.Compared with the negative control group,MYO6 expression was reduced in lentivirus infected SW1116 cells.3.Knockdown of MYO6 inhibited cell proliferation(P<0.001),restrained colony formation(P<0.001),and induced cell apoptosis(P<0.001)in SW1116 cells.Conclusions: Downregulation of myosin Ⅵ reduced cell growth and induced apoptosis in colorectal cancer. |
