| Objective: To investigate gene polymorphisms of leptin gene promoter region G2548 A,MMP-2(matrix metalloproteinase-2)gene C735 T,MMP-9(matrix metalloproteinase-9)gene C1562 T and pregnancy-induced hypertension syndrome(PIH)in Han population of Qinghai Province.PIH subjects(n=130)and normal pregnancy subjects(n=135)were studied to detect their serum level of leptin,MMP-2,MMP-9.The expression of leptin,MMP-2,MMP-9 m RNA in placenta tissue was also compared.Methods: Genomic DNA was extracted from blood.PCR-RFLP method was used to examine the leptin G2548 A,MMP-2 C735 T and MMP-9 C1562 T genotypes.The expression of leptin,MMP-2 and MMP-9 m RNA of placenta tissue were detected by RT-q PCR.Serum sample of Leptin,MMP-2,MMP-9 was tested by using ELISA.Results: 1.It was different of genotype frequency distribution of Leptin gene G2548 A.G allele might be the susceptible gene of PIH(OR = 1.361,95%CI: 1.01-1.83).2.It was different of genotype frequency distribution of MMP-2 gene C735 T.C allele might be the susceptible gene to PIH.(OR =1.53,95%CI: 1.07-2.20).3.The distribution of C allele and T allele of MMP-9 C1562 T have no difference between PIH group and control group(X2=2.40,P=0.12).4.The Leptin content of plasma(440.47±62.55)pg/ml in PIH group is higher than the control group(201.4±61.011)pg/ml,(P< 0.05).The MMP-2 content of plasma(24.95±4.80)pg/ml in PIH group is lower than the control group(37.81±4.40)pg/ml,(P< 0.05).The MMP-9 content of plasma(171.78±17.29)pg/ml in PIH group is lower than the control group(247.64 ±14.83)pg/ml,(P< 0.05).5.The expression of Leptin gene m RNA(3.41±0.97)in PIH group was higher than the control group(1.01±0.07),(P < 0.05).The expression ofMMP-2 gene m RNA(0.15±0.04)in PIH group was higher than the control group(1.02±0.05),(P< 0.05).There is no difference between the expression between MMP-9 gene m RNA(1.01±0.12)in the PIH group was higher than the control group(1.05±0.09),(P> 0.05).Conclusion: 1.Leptin gene G2548 A,MMP-2 gene C735 T polymorphisms are related to pregnancy induced hypertension disease of Han nationality in Qinghai province.G allele of Leptin gene G2548 A,C allele of MMP-2 gene C735 T were susceptible to pregnancy induced hypertension disease.MMP-9 C1562 T polymorphisms are not related to pregnancy induced hypertension disease of Han nationality in Qinghai province.2.The expression of Leptin,MMP-2 and MMP-9 are associated with the occurrence of pregnancy induced hypertension disease of Han nationality in Qinghai province. |
