Objective: Preliminarily screening plasma specific mi RNAs of two types of echinococcosis.Methods: Plasma samples were collected from 3 patients with alveolar echinococcosis(AE),3 patients with cystic echinococcosis(CE)and 3 healthy volunteers.The expression mi RNA profiles of three groups were detected by mi RNA microarray,and the differentially expressed mi RNAs between the each group were analyzed.The target genes of micro RNAs were predicted and analyzed by GO and KEGG pathway.Network relationships between differentially expressed micro RNAs and the key regulatory genes was constructed to screen out core micro RNAsResults: Compared with healthy volunteers,there were 144 differentially expressed micro RNAs in AE patients,69 of which are up-regulated,75 of which are downregulated;in CE patients,30 differentially expressed micro RNAs were screened out compared with healthy volunteers,12 of which are up-regulated,and 18 of which are down-regulated;17 micro RNAs expressed co-difference in AE patients and CE patients compared with healthy volunteers,4 of which are up-regulated,and 13 of which are down-regulated;and 21 differentially expressed micro RNAs in AE patients compared with and CE patients,1 of which is up-regulated,20 of whcih are downregulated.Bioinformatics analysis revealed that 4 kinds of mi RNAs(hsa-mi R-24-3p,hsa-mi R-328-3p,hsa-mi R-148b-3p,hsa-mi R-671-5p)were the key mi RNAs of two kinds of echinococcus to regulate the microenvironment.Seven kinds of mi RNAs(hsami R-17-5p,hsa-mi R-20b-5p,hsa-mi R-20a-5p,hsa-mi R-15a-5p,hsa-mi R-16-5p,hsami R-107,hsa-mi R-26b-5p)are the key factors in the survival and pathogenesis of alveolar echinococcus in human body Bonded micro RNAs;1 kind of mi RNAs(hsami R-96-5p)is the key mi RNAs for the survival and pathogenesis of cystic echinococcus in human body.Conclusion: Through miRNA microarray detection and bioinformatics analysis,we preliminarily screened out four kinds of micro RNAs with significant differences between patients and healthy volunteers,seven kinds of AE specific micro RNAs and one kind of CE specific micro RNAs.These micro RNAs are useful in early diagnosis of echinococcus,research on pathological mechanism of echinococcus and exploration of potential therapeutic targets. |
