Objective: To study the expression and clinical significance of PRRX1 and EMT related proteins in gastric cancer and metastatic lymph nodes tissues, and to analyze the correlation between PRRX1 and EMT related proteins, and to explore the relationship between the expression of PRRX1 and lymph node metastasis of gastric cancer. Methods: 93 cases of gastric cancer and 65 cases of metastatic lymph nodes were collected.the expression of PRRX1,E-cadherin and Vimentin in gastric cancer tissues and metastatic lymph nodes was detected by immunohistochemical SP method. To analyze the expression difference and correlation of PRRX1,E-cadherin and Vimentin in gastric cancer and metastatic lymph nodes, and their clinical significance. Results: 1.The positive rates of PRRX1 and Vimentin in gastric cancer(69.89% and 63.44%)were significantly higher than those in metastatic lymph nodes(43.08% and 36.92%)(P < 0.05).The positive rate of E-cadherin in gastric cancer(44.09%)was significantly lower than that in metastatic lymph node(61.54%)(P < 0.01);2.Expression of PRRX1 in metastatic lymph nodes and depth of invasion,TNM stage, differentiation, lymphatic invasion and blood vessels of gastric cancer.Invasion and nerve invasion were closely related(all p < 0 05).3.In metastatic lymph nodes, the expression of PRRX1 was positively correlated with Vimentin(r ≥ 0.429,p < 0.05)and negatively correlated with E-cadherin(r ≥ 0.334,P < 0.05).4.Logistic regression analysis showed that the expression of PRRX1,tumor size and depth of invasion were independent predictive factor for lymph node metastasis of gastric cancer. Conclusion: 1.PRRX1,E-cadherin and Vimentin were differentially expressed in gastric cancer tissues and metastatic lymph nodes, and relate to lymph node metastasis of gastric cancer.2.The transformation of EMT-MET may occur in the process of lymph node metastasis of gastric cancer.3.PRRX1 can be used as a new molecular marker for predicting lymph node metastasis of gastric cancer.
