General anesthesia is often required in animal experiments, and the choice of anesthetics and anesthesia methods have a large impact on the results. The most commonly used general anesthetics are sodium pentobarbital, chloral hydrate and urethane. The mechanism and the effect of each anesthetic are different. However, the effect of an esthetics in the cerebral cortex and specific functional areas is not very clear. With the widespread use of electronic manufacturing technology and the development of animal physics diagnostics technology, anesthesia testing has evolved from past damaging intermittent monitoring to non-invasive dynamic continuous monitoring.Photoacoustic tomography(PAT)is an emerging non-invasive biomedical imaging method and has become one of the fastest growing imaging methods in the field of biomedical optics.Unlike pure optical imaging(such as confocal microscopy and twophoton microscopy),PAT uses acoustically detected high optical absorption contrast and high resolution deep scattering tissue for imaging. In this paper,we explored the corresponding effects of different general anesthetics on the cortex by using this new biomedical imaging method, and combined with acupuncture to monitor the changes of cortical hemodynamics. In this paper, the experiment is based on photoacoustic tomography system, the effects of acupuncture on hemodynamics of mouse cerebral cortex under different anesthesia conditions were explored. The main research include:1.It mainly introduces the development history of general anesthesia and acupuncture and Zusanli in animal experiments Domestic and foreign, and expounds the importance of general anesthesia for modern medical research.2.It mainly introduces the basic principles and deficiencies of traditional biomedical imaging technology, the basic principles and classification of photoacoustic tomography, and the differences and advantages of photoacoustic tomography compared with traditional anesthesia monitoring imaging techniques.3.Introduced the laboratory photoacoustic tomography system, including the design of laboratory hardware configuration and control software, the construction principle of photoacoustic tomography experimental system, and the principle and algorithm of photoacoustic tomography, which is a smooth experiment. The theoretical foundation and experimental conditions were laid.4.Three animal anesthetics were used to study the hemodynamics of living brains based on photoacoustic tomography system. The differences in the effects of three different anesthetics on the functional areas of the cerebral cortex were investigated. At the same time, acupuncture and moxibustion were used to monitor the changes of hemodynamics in the cerebral cortex.PAT was used to study the relationship between the effects of general anesthesia on cerebral hemodynamics and vascular system in animal experiments and the concentration of total hemoglobin(HbT)in cortical vascular network, which promoted the study of PAT in general anesthesia and brain science, and also expanded PAT.In the range of biomedical applications.
