Single Nucleotide Polymorphisms In ZEB2 And The Effect Of Radiotherapy In Patients With Small Cell Lung Cancer

Objective: To explore the relationship between Single Nucleotide Polymorphisms(SNPs)in ZEB2 and the short-term effect of radiotherapy in patients with small cell lung cancer(SCLC).Methods: 121 patients with small cell lung cancer who accepted radiotherapy from September 2015 to October 2017 in the First Affiliated Hospital of China Medical University were selected.The genome DNA was extracted from blood before radiotherapy,MALDI-TOF MS was used to detect the SNPs in ZEB2 and The experimental data was analyzed with Typer4.0 software,the results were obtained.The RECIST 1.1 standard was used to evaluate the effect of radiotherapy,using chi-square(χ2)test and unconditional logistic regression model to analysis the relationship of the short-term effect of radiotherapy and SNPs genotypes and alleles in ZEB2.Results: 1.rs373398301,rs528611118,rs752715025 had no mutation in the target population and MAF in rs12105918 was less than 10%.2.rs6755392 genotypes and alleles were not related to the effect of radiotherapy(P > 0.05).3.rs3806475 alleles were associated with the radiotherapy effect in 1 month and 3 months to the end of radiotherapy(P < 0.05),and the patients who carrying AG+GG allele had better effect of radiotherapy than those who with AA allele in the medium term of radiotherapy,1 month and 3 months to the end of radiotherapy(OR=0.361 95%CI:0.138-0.940,OR=0.37395%CI:0.148-0.941 and OR=0.296 95%CI:0.116-0.755).4.rs3806475 genotypes were not related to the effect of radiotherapy in the medium term of radiotherapy(P > 0.05).The genotypes were related to the effect of radiotherapy in 1 month and 3 months after radiotherapy(P < 0.05),and the patients who carrying GG genotype had better effect of radiotherapy than those with AA genotype(OR=0.219 95%CI:0.058-0.832 and OR=0.142 95%CI:0.034-0.600).Conclusion: The rs3806475 in ZEB2 was associated with the short-term effect of radiotherapy in patients with small cell lung cancer,and the patients who carrying mutant type GG genotype had better short-term effect than those with the wild type AA genotype.
