Objective: The aim of the present study was to investigate the peripheral blood lymphocytes β3-adrenergic receptor(β3-AR)expression in patients with atrial fibrillation(AF)to analyze the predictive value of β3-AR expression in AF.Methods: 1.Research objects: A total of 98 patients with AF diagnosed clinically were selected as the AF group(35 patients with paroxysmal AF,33 patients with persistent AF and 30 patients with permanent AF)and 31 healthy adults were inrolled as control group.2.Research Method: Two groups of general clinical data and related biochemical parameters were collected.The lymphocytes were separated from peripheral blood samples of the research objects,and then total RNA was extracted from the lymphocytes,followed by the reverse transcription reaction for c DNA synthesis.After that,quantitative real-time PCR reaction was performed to detect the Ct values of lymphocytes β3-AR mRNA.The formula(objective gene Ct value-reference gene Ct values = △Ct values)was used to obtain △Ct values.Finally,the relative levels of lymphocytes β3-AR mRNA expression were calculated using the 2-△△Ct method.3.Primer designs and syntheses: primers of β3-AR mRNA were synthesized by Shanghai Sangon Biological Engineering Technology Services Reagents Co.,Ltd.4.All statistical analyses were conducted using SPSS22.0 software.Continuous variables were expressed as mean ± standard deviation(SD,`x±s)if they were normally distributed.The statistical comparison between two groups was performed using the two independent-samples t-test.If continuous variables were not normally distributed,they were made with the nonparametric Mann–Whitney U test.One-way analysis of variance(ANOVA)was used for multiple group comparisons.Groups for categorical variables were assessed by chi-square or Fisher’s exact tests.The correlation between the beta 3-AR and the different indexes was calculated by Pearson’s correlation analysis.Sensitivity and specificity were assessed by ROC curve analysis.A P value <0.05 was regarded as statistically significant.Results: 1.Compare to the control group,the expression level of β3-AR in AF group was significantly increased(P<0.05).2.The differences of expression level of β3-AR were statistically significant among patients with paroxysmal AF,persistent AF and permanent AF,and the longer extension of the AF duration,the higher the expression of β3-AR.3.Pearson correlation analysis showed that relative β3-AR mRNA expression was positively associated with left atrial diameter(LAD)in AF group(r=0.256,P<0.05).4.Binary stepwise logistic regression analysis(Backward selection)showed that LAD,CRP,β3-AR is closely correlated with AF.5.Receiver operating characteristic curves showed that the area under the curve of β3-AR was 0.734 in the prediction of AF(95%CI:0.639~0.829,P<0.05).Conclusions: 1.β3-AR expression is significantly elevated in patients with AF,and the longer extension of the AF duration,the higer the expression of β3-AR.It can be inferred that β3-AR may be the potential candidate biomarker of AF.2.β3-AR expression was positively associated with LAD in patients with AF,which may be involved in atrial structural remodeling.
