Objective: To study the expression of autophagy-related proteins LC3,Beclin-1,and mTOR in the marginal liver tissues and normal liver tissues of hepatic alveolar echinococcosis,and to compare to the difference of these proteins in the Hepatic alveolar echinococcosis,and to detect the relationship between autophagy and hepatic alveolar echinococcosis and its clinical significance.Methods: There were the marginal liver tissue of echinococcosis lesions and normal liver tissue collected from patients with hepatic alveolar echinococcosis from January to December 2018 in Affiliated Hospital of Qing Hai University.The patients included in the experiment underwent routine abdominal CT and MRI examinations,as well as laboratory indicators such as TB、ALB、ALT、AST、APTT and antibody IgG.The tissue specimen immunohistochemistry was used to detect the expression of LC3,Beclin-1 and mTOR in marginal liver tissue of echinococcosis lesions and normal liver tissue.The difference of expression of LC3,Beclin-1 and mTOR proteins and the correlation between LC3,Beclin-1 and mTOR protein expression in marginal liver tissue of echinococcosis lesions and normal liver tissue were compared.Correlation between protein expression and clinical pathological factors of patients: such as age,gender,maximum diameter of lesion,whether it is intrahepatic,metastasis,and TB、ALB、ALT、AST、APTT,etc were detected.Results: 1.The mean intensity of LC3 protein in marginal liver tissue of hepatic alveolar echinococcosis was significantly higher than mean intensity of LC3 protein in the normal liver tissue(P<0.05).2.The mean intensity of Beclin-1 protein was significantly higher than mean intensity of Beclin-1 protein in normal liver tissue,(P<0.05).3.The mean intensity of mTOR protein was lowly was significantly lower than the mean intensity of normal liver tissue(P<0.001).4.The mean intensity of LC3 protein expression was negatively correlated with mTOR protein expression(p<0.05),The mean intensity of Beclin-1 protein had no correlation with the mean intensity LC3,mTOR protein.5.The mean intensity of LC3 in the marginal liver tissue of hepatic alveolar echinococcosis was negatively correlated with TB(P<0.05),the mean intensity of LC3 has no correlation with anther indicators such as ALB、ALT、AST、APTT,ect.And The mean intensity of mTOR and Beclin1 in the marginal liver tissue of hepatic alveolar echinococcosis had no correlation with TB,ALB,ALT,AST,APTT,ect.And the mean intensity of LC3,mTOR in the all groups such as age,gender,lesion diameter,multiple lesions and single lesion,ect,have no difference.While the mean intensity of Beclin-1 protein in the group hepatic signal lesions was higher than the mean intensity of Beclin-1 protein in the group of hepatic multiple lesion(p<0.05),and the mean intensity of Beclin-1 protein in other groups have no difference.Conclusion: The results of the experiment indicated that: 1.The expression of autophagy-associated protein LC3 and Beclin-1 was significantly increased in the marginal liver tissue liver tissue of the HAE lesion.the mean intensity of mTOR in the marginal liver tissue of hepatic alveolar echinococcosis was significantly decreased,indicating that autophagy may play a role in the occurrence and development of hepatic alveolar echinococcosis.2.The low expression of Beclin-1 maybe related to the multiple infiltration mechanism of hepatic alveolar echinococcosis. |
