Objective Retrospectively analyze and summarize the medical records of patients with laryngeal squamous cell carcinoma archived in the Department of Otorhinolaryngology Head and Neck Surgery,Ningxia Medical University General Hospital for 10 years(January2009-December 2018),and detect laryngeal squamous cells Expression of DNA and its subtypes of human papillomavirus in cancer.To investigate whether HPV(Human Papillomavirus,HPV)infection is associated with the incidence of laryngeal squamous cell carcinoma in Ningxia.To lay a virological basis for the study of the etiology of laryngeal squamous cell carcinoma in the region,and to provide research directions for the prevention,diagnosis and treatment of HPV infection-associated laryngeal squamous cell carcinoma.Methods A total of 125 paraffin-embedded specimens from patients with laryngeal squamous cell carcinoma who were diagnosed as pathologically diagnosed laryngeal squamous cell carcinoma from January 2009 to March 2018 in Ningxia Medical University General Hospital were included in the experimental group.The clinical records of all enrolled patients were complete.The patients were all resident in Ningxia for 3 years and above;patients with laryngeal squamous cell carcinoma did not receive radiotherapy or chemotherapy before surgery;patients with vocal cord polyps had no history of malignant tumors of the head and neck.The clinical records of all enrolled patients were retrospectively analyzed,and the paraffin-embedded specimens of the selected patients were tested for HPV-DNA by in situ acid hybridization(In Situ Hybridization,ISH).?~2 test or the Fisher exact probability method.Using SPSS 19.0 software,the count data were statistically analyzed using the P(27)0.05 was considered statistically significant.Results(1)Clinical data analysis showed:experimental group:121 males and 4 females;aged 36-78 years,mean age 61.2±8.97 years;99 cases of smoking,31 cases of drinking;111cases of Han people,14 cases of ethnic minorities;according to age:4 young people(18-44years old),52 middle-aged(45-59 years old),69 elderly(60 years old and above);according to pathological classification:45 cases with high differentiation,52 cases with moderate differentiation,28 cases with poor differentiation According to the location of the disease:31cases of glottic type,90 cases of glottic type,4 cases of subglottic type;21 cases of lymph node metastasis.The control group:38 males and 3 females;aged 30-49 years,mean age42.3±2.75 years;(2)The total positive rate of HPV detection in the experimental group was71.2%;the positive rate of HPV16/18 subtype was 60.8%;the positive rate of HPV6/11subtype was 32.8%.The total positive rate of HPV detection in the control group was9.7%.The positive expression of HPV infection in laryngeal squamous cell carcinoma and vocal cord polyps was statistically significant(?~2=47.3,P(27)0.001);(3)There was no significant difference in HPV positive expression between patients with laryngeal squamous cell carcinoma in terms of gender,age,ethnicity,tumor differentiation,history of smoking or drinking,and lymph node metastasis(P(29)0.05).Conclusion(1)The male sex incidence of squamous cells in the throat of Ningxia is higher than that of females,the age is mainly concentrated in 60-78 years old,the smokers account for 79.2%,and the drinkers account for 24.8%;(2)the HPV infection rate in laryngeal squamous cell carcinoma is significantly higher than that.HPV infection rate in vocal cord polyps,HPV infection mainly HPV16/18 type,suggesting that HPV infection may be one of the important causes of laryngeal squamous cell carcinoma in Ningxia;(3)HPV infection rate of laryngeal squamous cell carcinoma in Ningxia is gender,age,ethnicity,lifestyle(smoking,drinking history,etc),degree of differentiation,There was no significant difference in lymph node metastasis.
