| Objective:To investigate the suppression effect of allicin combined 5-FU on MEC-1 xenografts tumer in nude mice and the expression levle of NF-κB p65,study the anti-tumor mechanism of allicin and provide experimental basis for the treatment of mucoepidermoid carcinoma.Methods:Put 24 male,26-30day BALB/c nude mice in SPF laboratory for one week.The cultured MEC-1 cells were adjusted to a concentration of 1×10 ~6/ml in PBS,subcutaneously inoculated with 0.2ml into the nude mouse left axilla to establish xenografting models.24 male nude mice were randomly grouped as model control group,5-FU group,allicin group and combination group by ear-cutting numbering method.The tumor formation,the size of Xenografted tumor,the morphology of the tumor with HE staining after tumor removal,detected the expression of NF-κB p65 by immunohistochemistry.Results:1.This study successfully established the MEC-1 xenograft model in nude mice.After 5days inoculation,24 nude mice all had tumor-like masses in subcutaneous tissue whose diameters were greater than 5mm and no mice died in the experiment.xenograft tumor transplantation rate was 100%.2.After drug intervention,measured the tumor volume of xenografted tumor,drawn the growth curve to understand the growth trend with time.Tumor volume increased with time after intervention and there was a difference in tumor volume between the four groups.The growth rate of 5-FU group,allicin group and combination group was significantly lower than that of the control group.3.The results of HE staining showed that compared with the tumor tissue of the model group,the tumor necrosis area of the allicin group,5-FU group and combination group greatly increased,and the most obvious group was the combination group.Compared with the control group,the relative expression of NF-κB p65 of other groups decreased,and the most obvious group was 5-FU combined with allicin(p<0.05).Conclusion:1.5-FU and allicin have inhibitory effects on MEC-1 in nude mice,and the effect is better when combined.2.5-FU and allicin can inhibit the expression of NF-κB p65 in tumor tissues,and the5-FU and allicin combined effect is more obvious. |
