Objective:Immunohistochemistry and Western blot were used to detect the expression of COX-2 and VEGF in colon adenocarcinoma,high-grade intraepithelial adenoma,low-grade intraepithelial adenoma and adjacent normal tissue In this topic.The correlation of CD105-labeled microvessel density,COX-2,VEGF protein and clinical pathological parameters of colon adenocarcinoma was analyzed by counting the number of microvessels(CD105-MVD)of each field of per visual field in different tissues using the CD105 marker.The purpose of these tests is to explore the role of COX-2,VEGF and microvessels in the development of colon adenocarcinoma,and to provide new ideas for early diagnosis and prognosis evaluation of colon adenocarcinoma.To explore the relationship between COX-2,VEGF expression and micro angiogenesis by expressing COX-2 and VEGF in colon cancer tissues and the correlation with CD105-MVD.This study provides new ideas for exploring the possible mechanisms of COX-2 and VEGF in the development of colonic adenocarcinoma.The study provides new ideas for studying the possible mechanisms of COX-2 and VEGF in the development of colon cancer.Methods:80 cases of colon adenocarcinoma,50 cases of high-grade intraepithelial neoplasms of colon gland epithelium,80 cases of low-grade intraepithelial neoplasms of colon gland epithelium,and 80 cases of adjacent normal tissue were collected from the surgical oncology of the First Central Hospital of Baoding City.The expression of COX-2,VEGF and CD105protein in each tissue were detected by immunohistochemical SP method,and CD105-MVD was counted.20 fresh colon adenocarcinoma specimens,20fresh adenoma specimens(high-grade epithelial neoplasia+low-grade epithelial neoplasia)and 20 fresh adjacent normal tissue were collected and preserved in-80℃refrigerator.The expression levels of COX-2 and VEGF proteins in various tissues were detected by Western blot.Results:1 Immunohistochemistry was used to detect the expression of CD105,COX-2and VEGF protein in colon tissues1.1 Immunohistochemistry was used to detect the protein expression of CD105 in colon tissuesThe CD105-MVD count was(32.10±5.10)in 80 cases of colon adenocarcinoma,(21.90±2.40)in high-grade intraepithelial neoplasms of colon gland epithelium,(23.50±5.30)in low-grade intraepithelial neoplasms of colon gland epithelium,and(12.30±2.80)in adjacent normal tissue.The difference was statistically significant among all groups,(P<0.05).1.2 Immunohistochemistry was used to detect the protein expression of COX-2 in colon tissuesIn 80 cases of colon adenocarcinoma,the positive rate of COX-2 protein expression was 58.75%(47/80).In 50 cases of high-grade intraepithelial neoplasms of colon gland epithelium,the positive rate of COX-2 protein expression was 30.00%(15/50).In 80 cases of low-grade intraepithelial neoplasms of colon gland epithelium,the positive rate of COX-2 protein expression was 28.75%(23/80).In 80 cases of adjacent normal tissue,the positive rate of COX-2 protein expression was 15.00%(12/80).The difference between the four groups was statistically significant,(χ~2=31.95,P<0.05).1.3 Immunohistochemistry was used to detect the protein expression of VEGF in colon tissuesIn 80 cases of colon adenocarcinoma,the positive rate of VEGF protein expression was 70.00%(56/80).In 50 cases of high-grade intraepithelial neoplasms of colon gland epithelium,the positive rate of COX-2 protein expression was 46.00%(23/50).In 80 cases of low-grade intraepithelial neoplasms of colon gland epithelium,the positive rate of COX-2 protein expression was 31.25%(25/80).In 80 cases of adjacent normal tissue,the positive rate of COX-2 protein expression was 21.25%(17/80).The difference between the four groups was statistically significant,(χ~2=40.46,P<0.05).2 Western blot method was used to detect the protein expression of COX-2and VEGF in colon tissues2.1 Western blot method was used to detect the protein expression of COX-2in colon tissuesThe relative expression of COX-2 protein in colon adenocarcinoma tissue was(4.20±0.81).The relative expression of COX-2 protein in colon adenoma was(3.09±0.87).The relative expression of COX-2 protein in adjacent normal tissue was(1.29±0.36).The difference between the three groups was statistically significant,P<0.05.2.2 Western blot method was used to detect the protein expression of VEGF in colon tissuesThe relative expression of VEGF protein in colon adenocarcinoma tissue was(3.27±0.85).The relative expression of COX-2 protein in colon adenoma was(1.22±0.58).The relative expression of COX-2 protein in adjacent normal tissue was(0.72±0.12).The difference between the three groups was statistically significant,P<0.05.3 The relationship between the expression of CD105,COX-2 and VEGF in colon cancer tissues and clinicopathological parameters3.1 The relationship between the expression of CD105 in colon cancer tissues and clinicopathological parametersCD105-MVD in colon adenocarcinoma was not correlated with age,sex,tumor length,lymph node metastasis and histological grade(P>0.05),but was correlated with the depth of tumor invasion(P<0.05).3.2 The relationship between the expression of COX-2 in colon cancer tissues and clinicopathological parameters.The expression of COX-2 protein in colon adenocarcinoma was not correlated with age,sex,tumor length and depth of invasion(P>0.05),but with histological grade and lymph node metastasis(P<0.05).3.3 The relationship between the expression of VEGF in colon cancer tissues and clinicopathological parameters.The expression of VEGF protein in colon adenocarcinoma was not correlated with age,sex,tumor length,histological grade and lymph node metastasis(P>0.05),but with the depth of invasion(P<0.05).4 Correlation between COX-2,VEGF expression and CD105-MVD in colon cancer tissues.The expression of COX-2 and VEGF protein in colon adenocarcinoma was positively correlated with CD105-MVD.Conclusion:1.The number of CD105-MVD in adjacent normal tissue,low-grade intraepithelial adenoma tissue,high-grade intraepithelial adenoma tissue and adenocarcinoma tissue showed an upward trend,and was related to the depth of invasion of tumor tissue.These suggested that microangiogenesis plays an important role in tumor invasion.2.The expression of COX-2 protein in adjacent normal tissue,low-grade intraepithelial adenoma tissue,high-grade intraepithelial adenoma tissue and colon adenocarcinoma tissue showed an upward trend,and correlated with histological grade and lymph node metastasis.These suggested that COX-2protein may be related to the differentiation and metastasis of colon tumors.3.The expression of VEGF protein in adjacent normal tissue,low-grade intraepithelial adenoma tissue,high-grade intraepithelial adenoma tissue and colon adenocarcinoma tissue showed an upward trend,and was related to the depth of invasion of tumor tissue.These suggested that VEGF protein may promote the occurrence and development of colon tumors.4.High expression of COX-2 and VEGF proteins in colon adenocarcinoma promote the occurrence and development of tumor,which may be related to the formation of tumor microvasculature.
