| The onset of T2DM is related to the dysfunction of the HPA axis,characterized by abnormal glucose and lipid metabolism and insulin resistance.At present,the treatment of T2DM,the use of oral hypoglycemic drugs or insulin injections to maintain blood sugar levels,and the treatment of traditional Chinese medicine more focused on regulating insulin resistance,but neglected the neuroendocrine on the occurrence of T2DM,the role of development.Therefore,the study of the sculellaria barbata and the HPA axis of diabetic rats as carrier,using COD-PAP method,GPO-PAP method,direct determination method,HE staining,PAS staining,aldehyde complex red staining method,immunohistochemical SABC method are used.to elucidate the relationship between the morphology and function of HPA axis in T2DM rats and glycolipid metabolism and the therapeutic effect on T2DM under the intervention of scutellaria barbata.Experiment one.Establishment of T2DM ModelThe selection of Wistar SPF rats 168,4 weeks of age,male,according to the principle of no difference in weight randomly divided into health control group(48)and model group(120).All rats are fed with normal feedstuffs for 1 week and model group rats are changed to high-sugar high-fat diet and fed continuously for 30 days.31 d,24-hour fasting.In experiment 32 d,all rats are weighed and the model started.During the modeling process,the model group was used an intraperitoneal injection of 5‰STZ at a body weight of 45mg/kg,and the health control group was intraperitoneally injected with a citrate buffer of 4.5ml/kg for 30 min.In experiment 34 d,38d,45 d and 59 d,the body weight and water intake of the rats are observed;the FBG changes are measured by blood sampling;the changes of INS,CORT,TC,TG,HDL-C,and LDL-C are measured by serum;the hypothalamic PVN and VMH,adenohypophysis,adrenal glands and pancreas are observed pathological changes by HE staining.PAS staining was used to observe the change of the number of glycogen in hypothalamic PVN and VMH,adenohypophysis,and adrenal glands.The pancreatic α cells,β cells,and collagen fibers are observed by aldehyde-red staining method,immunohistochemical SABC method to observe the expression of 5-HT and GABA in the PVN and VMH.The experiment results are as follows: Compared with the healthy group,the model group had higher drinking water and lower body weight.The model group fasting blood glucose increased.The contents of INS in serum are decreased.In the serum,the contents of CORT,TC,TG,LDL-C increased.The contents of HDL-C droped in the serum.The number of neurons decreased,glial cells increased in hypothalamic PVN and VMH.The number of basophils decreased in the adenohypophysis.The adrenal band cells decreased and necrosis occurred.The number of pancreatic α and β cells decreased,necrosis occurred,and collagen fibers increased.In scutellaria barbata treated rats,the number of glycogen in the hypothalamus PVN,adenohypophysis and zona fasciculate cell decreased,and the amount of glycogen in the hypothalamus VMH increased.The expression of 5-HT and GABA increased in hypothalamic PVN large cells,and its expression in small cells decreased.The expression of 5-HT in the VMH region of excitatory neurons was reduced and high in the inhibitory neuron region.The GABA decreased in both glucose excitatory neurons and inhibitory neurons in the VMH.Experiment two.Effect of Sculellaria Barbata on T2DM and related indexes.On experiment 38 d determined the success of the model.All experimental animals are divided into 5 groups,which are healthy control group,model control group,scutellaria barbata low,middle and high dose groups,which are 24 rats in each group.On experiment 39 d,24-hour fasting,the experimental rats are given intragastric administration at regular intervals.On experiment 40 d,the scutellaria barbata low,medium and high dose groups are fed with different concentrations(0.1g/ml,0.25g/ml,0.5g/ml)of the extract according to the amount of 1ml/100 g for 30 days.The healthy control group and model group to the stomach to give 1ml/100 g saline.The above related indicators are observed on experiment 44 d,49d,59 d,69d,(scutellaria barbata treatment’s 5d,10 d,20d,30d).During the whole treatment process,the healthy control group was fed with basic feed,and the rest of the groups are fed with high-sugar high-fat diet,the experiment period of 30 d.The experimental results are as follows: Compared with the model control group,the weight of the rats increased in the scutellaria barbata treatment group,the FBG decreased,the CORT,TC,TG,LDL-C and INS decreased in serum,and the HDL-C increased in serum.The number of neurons increased and glial cells decreased in rat PVN and VMH.The number of basophils ncreased in the adenohypophysis.The adrenal zona fasciculate cells increased.The number of beta cells increased and collagen fibers decreased in the pancreas.The number of glycogen in PVN,adenohypophysis and adrenal zona fasciculate decreased,while the number of glycogen in VMH increased.The expression of 5-HT in the PVN of rats with the low and high doses was close to the healthy control group in treatment of 10 d,20d,30 d with the sculellaria barbata.In the treatment of 5d,10 d,the expression of excitatory neuron region and inhibitory nerve region of 5-HT in the low dose group of the rat VMH was tending to healthy control group.In In the 5th and 10 th day of scutellaria barbata treatment,the GABA expression levels in the VMH-GE region of the Scutellaria barbata low and high dose groups are similar to those in the healthy control group.In the treatment of 5d,10 d,the expression of GABA in the PVN of in low dose group was close to the healthy control group.The GABA expression levels in the GI region of the middle and high dose groups are similar to those in the healthy control group.GABA expression levels in the VMH-GE region of the scutellaria barbata low and high dose groups was similar to those in the healthy control group.The GABA expression levels in the GI region of the middle and high dose groups are similar to those in the healthy control group in the 5th and 10 th day of scutellaria barbata treatment.The results of the study show that: HPA axis participates in the pathogenesis of T2DM.The sculellaria barbata had an effect on abnormal glucose and lipid metabolism in T2DM rats.The sculellaria barbata had a positive effect on the function of HPA axis in T2DM rats.The effect of T2DM on hypoglycemic action and protective effect of HPA axis in rats had time and dose dependence by scutellaria barbata treatment.In the early stage of treatment(5d~10d),the effect of low and middle dose of scutellaria barbata was better.In the later stage of treatment(20~30d),the treatment of T2DM was better in the medium and high dose groups. |
