| Objectives:Inflammatory reaction and scar formation after peripheral nerve injury are the main factors that hinder nerve regeneration.And saikosaponin a has been shown to play a role in inhibiting inflammatory response in rats with spinal cord injury and brain injury.In this study,saikosaponin a was used in rats with sciatic nerve injury to study its effects on inflammatory response and nerve scar formation.At the same time neurophysiological and sciatic nerve function index were used to evaluate the recovery of neurological function.Methods:1.Rat models of sciatic nerve injury were established and divided into three groups: sham group(sciatic nerve was only exposed),SNI+SSa group(injection of saikosaponin a suspension after sciatic nerve was cut,dose 10 mg/kg),SNI group(intraperitoneal injection of the same amount of normal saline after cutting the sciatic nerve).2.Eight rats in each group were taken 7 days after model establishment,and the corresponding sciatic nerve was dissected and the expression level of anti-inflammatory factor IL-10 was detected by ELISA.3.The sciatic nerve function index of each group was measured at 1 day,7 days,14 days,28 days and 56 days after model establishment,and the recovery of lower limb motor function was evaluated.The neurophysiological function of each group was measured 56 days after modeling,and the recovery of nerve conduction function was evaluated.4.Six rats in each group were taken 56 days after modeling,and the corresponding sciatic nerve was dissected and measure the formation of scar tissue at the injury site by Masson staining.Meanwhile.At the same time,six rats in each group were taken 56 days after model establishment,and the corresponding sciatic nerve tissues were dissected and detect the content of type I and type III collagen by Western blot after treatment.Results:1.The results of ELISA detection of IL-10 expression in each group indicated that saikosaponin a promoted the expression of IL-10 in rats with sciatic nerve injury.The expression of IL-10 in sham group was 65.93±7.374ng/L,the expression level was 95.74±5.741ng/L in SNI group,and the expression level was 168.2±12.13ng/L in SNI+SSa group.2.Masson staining showed that the amount of blue collagen fibers in the sham group was small,and the blue collagen fibers in the injury group were abundant,while the amount of blue collagen fibers in the treatment group was significantly less than that in the injury group.Western blot results showed that the expression of type I collagen and type III collagen in the SNI group was significantly higher than that in the SNI+SSa group,and the difference was statistically significant.The above results together indicate that saikosaponin a inhibits the formation of scar tissue in the rat sciatic nerve injury.3.The measurement results of sciatic nerve function index showed that the SFI values of the SNI+SSa group were significantly higher than that of the SNI group at 28 and 56 days after model establishment,and the difference was statistically significant.The results of neuroelectrophysiology showed that the conduction velocity of sciatic nerve in the SNI+SSa group was significantly higher than that in the SNI group,and the difference was statistically significant.The above results indicated that saikosaponin a promoted the recovery of lower limb motor function and nerve conduction function in rats with sciatic nerve injury.Conclusions:1.Saikosaponin a can act on rat sciatic nerve injury by promoting the expression of IL-10 and inhibiting the formation of nerve scar.2.Saikosaponin a has a positive effect on lower limb motor function and nerve electrical conduction recovery after sciatic nerve injury in rats.Or it can be an aid in the treatment of peripheral nerve injury. |
