Objective:The PHF protein family is primarily a zinc finger protein containing a PHD(Plant Homeodomain)domain.Zinc finger protein 20(PHF20),a plant homologous domain,is a potential transcription factor that specifically recognizes methylated lysine and is a subunit of the lysine acetyltransferase complex MOF-NSL.Base,catalyzing histone H4 acetylation and non-histone acetylation.Studies have shown that the expression of PHF20 is closely related to the occurrence and development of breast cancer,but its molecular mechanism is not very clear.Therefore,this study aimed to preliminarily analyze the interacting proteins of PHF20 and initially explore the role of PHF20 in tumorigenesis and development.Methods:1.The proteins that may interact with PHF20 were identified by immunoaffinity purification combined with silver staining mass spectrometry.The results of immunostaining were used to verify the results of silver staining mass spectrometry.2.KEGG analysis,PPI analysis and GO analysis of the interacting proteins obtained by mass spectrometry to find the signaling pathways involved in epigenetic transcriptional regulation involved in PHF20 interacting proteins.3.In vitro experiments using breast cancer cell lines,including EDU for cell proliferation and Transwell assays for cell invasion.4.Further explore the association between PHF20 and malignant tumors using TCGA database,GEO database,GEPIA analysis and KM-plot survival analysis.Results:1.PHF20 can specifically bind to KDM4 A,EZH2,EED and Rb Ap46/48 to exert epigenetic regulation.2.PHF20 interacting proteins are involved in many epigenetic transcriptional regulation-related signaling pathways,such as chromatin assembly,histone modification,gene expression regulation,etc.The PHF20 interaction network contains many epigenetic-related complexes,such as Nu RD complexes,Pc G complex,MLL complex and DNA damage repair compound,etc.3.Ed U and transfer chamber experiments showed that the proliferation and invasion ability of PHF20 was significantly reduced compared with control cells.4.Clinical data analysis indicates that PHF20 is highly expressed in a variety of tumor tissues and is associated with poor prognosis.Conclusion:Our study was the first to discover the ability of PHF20 to interact with a variety of epigenetic regulatory complexes in vivo and to promote tumor cell proliferation and invasion.It is first discovered that PHF20 is highly expressed in breast cancer and various tumor tissues,suggesting that PHF20 may be a potential tumor marker,providing a new target for the diagnosis and treatment of breast cancer and other tumors.
