Erding Granule is a prescription recorded in Pharmacopoeia.It mainly consists of purple flower,dandelion,Radix Isatidis and Lobelia chinensis.It is clinically used as a heat-clearing and detoxifying agent.It is used for heat boil poison,throat swelling and pain,wind-heat and fire eye caused by heat and toxicity.It can be used for the treatment of inflammatory diseases[1]such as chronic bronchitis,acute pharyngitis.The research group and Sichuan Daqian Pharmaceutical Co.,Ltd.cooperated in the development project of"Erding Granule Increasing Function"and found that Erding Granule has significant effect on reducing uric acid,but its specific mechanism is still unclear.Therefore,based on the needs of the implementation of the cooperative project,the mechanism of Erding Granule in the intervention of hyperuricemia was studied in this paper,which includes the following aspects:1.By using potassium oxalate as modelling agent to establish hyperuricemia mice model,the pharmacodynamics of Erding granule water extract,50%ethanol extract and 95%ethanol extract of Erding granule were evaluated,and the best effective part of Erding granule for reducing uric acid was screened.The results showed that 50%ethanol extract from different extracts of Erding Granule had obvious hypouric acid effect.This laid a foundation for the further study of Erding Granule.2.Establishing a rat model of hyperuricemia by intraperitoneal injection of potassium oxalate and intragastric administration of hypoxanthine,the hypouric acid-lowering effect of the effective part of Erding Granule was studied.The pharmacodynamic evaluation of the effective part of Erding Granule was carried out by combining detection of relevant serum metabolic indicators,inflammatory factors and HE staining.The results showed that the effective parts of Erding Granule could improve the pathological damage of rat kidney tissue,inhibit the expression of inflammatory factors TNF-alpha,IL-1beta and IL-6,and the effect of reducing uric acid showed a dose-effect relationship.3.The expression levels of GLUT9 and OAT1 uric acid transporters were detected by Western biotting and real-time fluorescence quantitative PCR(q RT-PCR),and the mechanism of the effective fraction of Erding Granule in preventing and treating hyperuricemia model rats was explored.The results showed that the active fraction of Erding Granule could significantly up-regulate the expression of OAT1 and GLUT9 protein.The mechanism of reducing uric acid by the effective fraction of Erding Granule was verified at the molecular level,which provided a theoretical basis for expanding the clinical application of Erding Granule.In conclusion,the 50%ethanol extract of Erding Granule has the effect of reducing uric acid.It can prevent and treat hyperuricemia by improving renal pathological injury,inhibiting the expression of inflammatory factors TNF-α,IL-1β,IL-6 and regulating the expression of GLUT9 and OAT1. |
