| Objective:To observe the effect of Modified Wendan Tang on nutritional obesity in male rats and to explore its intervention mechanism in order to guide the clinical scientific weight loss.Method: Male SD rats of SPF grade were randomly divided into normal control group,(NC),model control group,(MC),modified Wendan Tang high dose group,(MHC),modified Wendan Tang medium dose group,(MMC),modified Wendan Tang low dose group,(MLC),Twenty in each group.Every group were given high-fat emulsion to establish nutritional obesity rats model,except for the normal control group.The rats in the constructed high-fat emulsion group were disrupted and re-grouped after 2 weeks and given corresponding drugs for 5 weeks in succession,during which body weight was measured regularly.When the last administration was finished,the objects in each group were fasted for 12 hours,within which water is allowed,then we opened their abdomen and separated the sera from the blood sampled via abdominal artery to detect the contents of TG、TC、GLU、LEP、ADP、INS、GC after anesthetizing the rats and measuring theirs length.The adipose tissue around viscera was isolated and the changes of pathological sections were observed.The contents of AMPK β,ACC and COX4 in the same part of rat liver and gastrocnemius muscle were detected by Elisa kit.Results: 1.Compared with the model control group,the modified Wendan Tang groups decreased Lee’s index and BMI,High dose was statistically significant(P < 0.05).2.Modified Wendan Tang groups decreased rats’ serum TG and TC compared with the model control group and the difference was not significant(P > 0.05).Compared with the model control group,the high and low dose of Modified Wendan Tang decreased the serum GLU,of rats,but the middle dose of modified Wendan decoction increased the serum GLU of rats,but the difference was not significant(P > 0.05).3.Compared with the model control group,the high dosage of modified Wendan Tang group reduced serum LEP,but it could increase LEP with the decrease of dosage and the difference was not significant(P > 0.05),all the three dosages could increase serum ADP content but the medium-dose had the most significant difference(P < 0.01).4.Compared with the model control group,all modified Wendan Tang groups could reduce serum INS(high and low dose have remarkably reduction)and increase serum GC(no significantly difference,P<0.01),high and low dose have remarkably reduction on serum INS(P>0.05).5.Compared with the model control group,all modified Wendan Tang groups could reduce AMPKβ,ACC and COX4 in gastrocnemius muscle tissue of rats,among which low-dose had huge affection on AMPKβ、ACC(p<0.01、p<0.05),so was blank group on ACC too,other than these two no big difference had shown.6.Compared with model control group,medium-dose modified Gallbladder-Warming Decoction group could increase AMPKβ hepatic tissue in rats and the other two doses had the opposite effect,though the difference was not significant(p > 0.05).In contrast to low dose’s reduction of ACC content in the liver tissue of rat,high dose and medium dose increased it(p>0.05).All doses could increase the content of COX4 with no significantly different(p > 0.05).7.Compared with the model control group,pathological sections showed that the modified Wendan Tang groups could reduce both the volume of Adipocytes significantly and the fatty change of hepatocytes.Conclusion: The oral administration of Modified Wendan Tang can resist the nutritional obesity of male rats and adjust the lipid metabolism disorder in rats.This may be achieved by improving the serum insulin resistance of the rat,raising the serum adiponectin in the rat,and participating in the regulation of the AMPK-ACC signal pathway associated with the lipid metabolism. |
