Objective"Ultra-high performance liquid chromatography-quadrupole-time-of-flight mass spectrometry and GAS chromatography-mass spectrometry were used to analyze the total components of Fructus Aurantii.Meanwhile,the gastric motility pharmacodynamic index of Fructus Aurantii was determined.Partial least squares(OPLS)was used to analyze the correlation between the components of Fructus Aurantii and pharmacodynamic index.The main active components of Fructus Aurantii closely related to GAS,VIP and 5-HT indexes were screened.The material basis of Fructus Aurantii for promoting gastrointestinal motility was discussed.Combined with pharmacopoeia index components,the equivalence of integrated decoction pieces and traditional decoction pieces was determined.Method(1)Ultra-high performance liquid chromatography-quadrupole-time of flight mass spectrometry(UPLC-Q-TOF-MS/MS)and gas chromatography-mass spectrometry(GC-MS)were used to analyze the total components of Aurantii Fructus.A total of 42 compounds were identified from the alcohol extract of Aurantii Fructus in positive ion mode.The structures of naringenin,5,6-dihydroxy-7,4’-dimethylflavone,limonin,5-norkawakacin,rutinaringin were analyzed and 22 volatile oil components of Aurantii Fructus were identified.(2)GAS,VIP and 5-HT were measured in mice with functional dyspepsia.Aurantii Fructus administration group significantly increased gastrin and 5-HT in model mice(P < 0.05)and significantly decreased VIP in model mice(P < 0.05).(3)Orthogonal Partial Least Squares(OPLS)was used to find the active components related to each pharmacodynamic index.The results showed that the possible active components of VIP index were loniceraside,5,6-dihydroxy-7,4’-dimethoxyflavone and naringin-7-O-glucoside.The possible active components of gastrin index are grape lactone,umbelliferone,Malmin,eriodictyol-7-O-glucoside,eriodictyol,and 5-nortangerin.The possible active components of 5-HT index are isosakurin,limonin,citron glycoside and epoxyhesperetin.(4)The contents of the components of the integrated decoction pieces and the traditional decoction pieces from the same origin were compared.First,there was no significant difference in the fingerprint compounds through PCA-X analysis.Then,the contents of naringin and neohesperidin,the index components of pharmacopoeia,were compared,and no significant difference was found.At the same time,the gastrin,VIP,5-HT and hemorheology of the integrated decoction pieces from the same origin and the traditional decoction pieces are compared.The results show that there is no significant difference in gastrin,VIP,5-HT and hemorheology indexes between different decoction pieces from the same origin.ConclusionsIn this paper,Aurantii Fructus was separated and identified by LC-MS and GC-MS,the chemical composition of Aurantii Fructus was comprehensively analyzed,and gastrointestinal motility indexes of model mice,such as gastrin,VIP and 5-HT,were determined,which confirmed that Aurantii Fructus has the function of promoting gastrointestinal motility.Then partial least squares(OPLS)was used to analyze the components related to the pharmacological effects of Aurantii Fructus.Possible active components closely related to each pharmacological effect were obtained,and the material basis of each pharmacological effect of Aurantii Fructus was preliminarily clarified.By comparing the components and drug effects of Aurantii Fructus from the same origin,it is determined that the integrated decoction pieces of Aurantii Fructus are equivalent to the traditional decoction pieces,which provides scientific basis for the quality evaluation standard and clinical application of Aurantii Fructus.
