Objective: To test the expression of GINS2 gene in malignant melanoma(MM),construct a GINS2-sh RNA lentiviral expression vector and identify it,and provide evidence for gene-targeted therapy of MM.Method:1.Immunohistochemical staining was used to analyze the expression of GINS2 in human malignant melanoma and adjacent tissues.q PCR was used to detect human invasive choroidal melanoma cell line(MUM-2C)and human malignant melanoma cell line(A375).GINS2 gene expression;2.The synthetic GINS2-shRNA sequence is ligated to the lentiviral vector GV115 for lentiviral packaging and quality assessment;3.The A375 cell line was transfected with GINS2-sh RNA lentivirus.The expression of GINS2 m RNA in sh GINS2 group and negative control group was detected by q PCR.The expression level of GINS2 protein in sh GINS2 group and negative control group was detected by Western-blot.Dyeing effect.Result:1.Immunohistochemical staining showed that the expression of GINS2 in MM tissue was significantly higher than that in adjacent tissues(P<0.05).q PCR detection showed that GINS2 was in MUM-2C cells and A375 cells.High abundance expression(ΔCt value ≤ 12).2.Successfully synthesized and identified GINS2-sh RNA lentiviral expression vector.3.The expression of GINS2 m RNA in sh GINS2 group was significantly lower than that in sh Ctrl group(P<0.01).The difference was significant in the sh GINS2 group.The GINS2 protein content was significantly lower than the sh Ctrl group.Conclusion:1.The expression of GINS2 gene in MM tissues and cells was significantly increased;2.The GINS2-sh RNA lentiviral expression vector was successfully constructed and successfully transfected into A375 cell line,which significantly decreased the m RNA and protein expression of GINS2 gene.It was confirmed that the GINS2-sh RNA recombinant lentiviral vector was successfully constructed.Lay the foundation for the role of MM development. |
