Objective In this experiment,Lewis lung cancer cells were used to set an model of non-small cell lung cancer in the mouse,and to evaluate the combination effect of anti-mouse-PD-1 monoclonal antibody and zoledronic acid(ZoL)to treat tumorbearing mice.Method Sixty C57BL/6 mice were randomly divided into six groups,the control group were injected PBS,and the Control anti-mouse Ab group,anti-mousePD-1,ZoL,ZoL+Control anti-mouse Ab,ZoL+ anti-mouse Ab.The anti-mouse-PD-1 was administered by tail vein injection once a week for 3 weeks,ZoL was administered intraperitoneally and administered continuously for 3 weeks,5 days a/week.In the living state,the peripheral blood of the inguinal vein was taken from the mouse.The expression of Pro-MMP-9 and VEGF in the serum after centrifugation and the immunohistochemical staining of the tumor tissue were used to evaluate the effect of ZoL on macrophages.Effect of anti-mouse-PD-1 on macrophages in tumor tissues.Results(1)The tumor suppression effect of anti-PD-1 monoclonal antibody group and ZoL + anti-PD-1 group was the most obvious,and there was no significant difference between the two groups(P >0.05).(2)ELISA results showed that the ProMMP-9 and VEGF levels in the a-PD-1 group,ZoL group and ZoL+a-PD-1 group were significantly lower than those in the PBS control group and Control Ab group with significant differences(P < 0.05).(3)Immunohistochemistry results showed ZoL group,and ZoL+ Control Ab drug There was a significant difference between the group and the control group(P<0.05).There was a significant difference between the ZoL group and the a-PD-1 group(P < 0.05).Conclusions(1)The combination of ZoL+anti-mouse-PD-1 and the anti-mouse-PD-1 monoclonal antibody had the same anti-tumor effect,which did not reach the experimental expectation.(2)The antimouse-PD-1 monoclonal antibody increased the number of macrophages in tumor tissues to some extent.
