Objective:Papillary thyroid carcinoma(PTC)is a common malignant tumor in the head and neck,and its incidence is increasing in recent years.Therefore,it is very important for us to explore suitable tumor markers for diagnosis and treatment of PTC.In this study,the expression of midkine protein(MK)and vascular endothelial growthfactor(VEGF)in papillary thyroid carcinoma tissues,adjacent normal tissues and nodular goiter tissues was detected by immunohistochemistry.Aim to investigate the correlation of MK and VEGF with clinical features of papillary thyroid carcinoma.Methods:The clinical data of patients undergoing thyroidectomy in otorhinolaryngology head and neck surgery in Shanxian central hospital from October 2015 to September 2016 were retrospectively studied.The paraffin specimens were collected and divided into three groups:experimental group(papillary thyroid carcinoma group,referred to as the cancer group)40cases,control group 1(paired normal thyroid tissues,referred to as adjacent to the cancer group)40 cases and control group 2(nodular goiter,referred to as the node group)40 cases.In this study,the expressions of MK and VEGF in three groups of thyroid tissues were studied by immunohistochemistry.The data were analyzed by SPSS 22.0 software.The positive expression rates of MK and VEGF and their relationship with the clinical features(including gender,age,lymph node metastasistumor,TNM staging and tumor diameter)of PTC were analyzed in three groups of thyroid tissues.Results:(1)The expressions of MK and VEGF proteins in the PTC were obviously stronger than that in the corresponding normal thyroid tissues and nodular goiter tissues(P < 0.05);(2)The expressions of MK and VEGF proteins in the PTC were correlated with lymph node metastasis(P<0.05),and the expressions of the two proteins were higher in the lymphatic metastasis group;(3)The expressions of MK and VEGF proteins had no significance with sex and age in PTC(P>0.05);(4)The expression of MK proteins in the papillary thyroid carcinoma were correlated with the diameter of tumor(P<0.05).The positive rate of MK in > 1cm group was higher than that in diameter≤1cm group.The expression of VEGF proteins in the PTC weren’t correlated with the diameter of tumor(P>0.05);(5)There was a positive correlation between the expression of MK and VEGF protein in PTC(r = 0.339,P<0.05).Conclusion:(1)The positive expression rate of MK and VEGF in PTC group was significantlyhigher than that in the paired normal thyroid group and nodular goiter group,and the difference was significant;(2)The positive expression of MK and VEGF correlated with lymphatic metastasis and TNM stages,but not with the gender and age of patients with PTC.(3)The combined detection of two protein markers,MK and VEGF,may play an important role in the diagnosis and prognosis of PTC.
