| ObjectiveIrisin is a new kind of cellular factor that is linked to energy metabolism,insulin resistance,etc,which is widely expressed in human body and closely related to the occurrence,development,invasion and migration of various tumors.However,there is currently little research on the biological effects of Irisin on epithelial ovarian cancer cells.This study aimed to clarify the regulation of Irisin on the cell cycle and the basic biological effects in the epithelial ovarian cancer cells.MethodsTo treat A2780 epithelial ovarian cells with different concentrations of Irisin.And its effects on cell growth、proliferation、cell cycle and cell cycle-related protein were respectively investigated by CCK-8 assay、flow cytometry and Western blot.Relevant statistical software was used to analyze and draw relevant conclusions.Results1.CCK-8 test:In the experimental group with Irisin concentration of 0.625,1.25,2.5,5 and10nmol/L,the A value of A2780 cells was 0.428±0.045、0.442±0.048、0.472±0.031、0.469±0.042、0.468±0.027,respectively.Compared with the A value of 0.381±0.030 in the control group(i.e,Irisin concentration of 0),the P<0.01,was statistically significant.Select the time curve(0h,12h,24h,36h,48h)for A2780 cells with Irisin concentration of 2.5nmol/L,with A values of 0.144±0.005、1.806±0.053、2.034±0.057、2.258±0.059、2.250±0.085,respectively.The A values of the same time in the control group(ie,the concentration of Irisin was 0)were 0.141±0.007、1.374±0.043、1.610±0.054、1.735±0.098、1.777±0.097,respectively.The difference was statistically significant in the experimental group of 12h、24h、36h and 48h compared with the control group.The P<0.01.2.Flow cytometry test:In A2780 cells which were not added to Irisin,about(59.3±5)%cells were accumulated in G0-G1 period,about(18.2±4)%cells,(22.5±6)%cells were accumulated in the period S、G2-M,respectively,in the cell cycle.In A2780 cells cultured with 2.5nmol/L Irisin,about(47.2±6)%cells were accumulated in G0-G1 stage,about(26.7±7)%cells,(26.1±4)%cells were respectively accumulated in the period of S,G2-M period,in the cell cycle.Compared to the group of no Irisin respectively,P<0.01,statistically significant.3.Western blot experiment was used to detect the expression of Cyclin D1,a protein related to cell cycle.Compared with the group of no Irisin,the relative expression level of Cyclin D1in the experimental group was(1.340±0.024).There’s a statistical significance to the difference between the two groups,and the t=19.63,P<0.0001.The expression level of Cyclin D1 in the experimental group was significantly raised.Conclusion1.Irisin can promote the growth and proliferation of epithelial ovarian cancer A2780 cells in a time-dependent manner.2.Irisin can accelerate the process of cell cycle and promote cells from G1 phase to S phase in the epithelial ovarian cancer A2780 cells.3.Irisin can upregulate the expression of Cyclin D1 in the epithelial ovarian cancer A2780cells. |
