Objectives The expression of CTNNB1 and LEF1 was detected at the level of esophageal scalar cancer cells and pathological tissues.The relationship between CTNB1and LEF1 and the radiation resistance of esophageal squamous carcinoma was discussed based on the difference of cell radiation resistance and the therapeutic effect of radiotherapy in patients.Methods The human esophageal scale cancer cell line TE13(radiosensitive cell line)and TE13R(corresponding radiosensitive cell line)4Gy dose were irradiated.The total RNA in TE13 and TE13R cells was extracted before(0h)and after irradiation(24h).The RT-PCR method verifies the expression of CTNNB1 and LEF1.The independent sample t test was used to compare the difference in mean number between the two groups.Paraffin pathological specimens of 59 patients with esophageal squamous carcinoma who received radical radiotherapy at tangshan people’s hospital were collected.According to the recent curative effect of radiotherapy in patients,they were divided into effective group and ineffective group.The expression of CTNNB1 and LEF1 in the pathological tissues of esophageal cancer patients was detected by immunohistochemical method.The difference of protein expression between the two groups was compared withχ~2 test.Results Cell experimental results confirmed that the expression of CTNNB1 in the radiation resistance cell line(TE13R)after exposure to radiation was higher than that of the radiosensitive cell line(TE13),and the difference was significant(P=0.016);The expression of LEF1 in the radiation resistance cell line(TE13R)after exposure to radiation was higher than that of the radiosensitive cell line(TE13),and the difference was significant(P=0.045).Tissue experiments confirmed that CTNNB1 protein is mainly located in cell membrane,cytoplasm and nucleus in esophageal squamous carcinoma.The difference in the overall expression level of CTNNB1 protein in effective group and ineffective group of radiotherapy did not have significant(χ2=0.827,P=0.363);The expression level of CTNNB1 protein nucleus was higher in the invalid group than in the effective group,and the difference was significant(χ2=4.911,P=0.027).The LEF1protein is mainly located in the cell nucleus.The LEF1 protein nucleus expression level was higher than that of the effective group,and the difference was significant in the invalid group(χ2=6.110,P=0.013).The results of Spearman correlation analysis indicate that there is a positive correlation between the expression of nuclear CTNNB1 protein and the expression of LEF1 protein in the pathological tissues of esophageal squamous carcinoma,rs=0.809,P<0.001.Conclusions The high expression of CTNNB1 may be related to the radiation resistance of esophageal squamous carcinoma,and CTNB1 may be an important indicator for predicting the radiation resistance of esophageal carcinoma.2 The high expression of LEF1 may be related to the radiotherapy resistance of esophageal squamous cancer,and LEF1 may be an important indicator for predicting the radiation resistance of esophageal cancer.The nuclear expression level of CTNNB1 in.3 esophageal squamous carcinoma tissue was positively correlated with the LEF1 expression level,and may be related to the activation process of Wnt pathway.4 Wnt pathway activation may be one of the molecular molecular mechanisms for the formation of radiation resistance to esophageal cancer.Figure 5;Table 6;Reference88. |
