| Objectives In order to further clarify the effect of CIH on the expression of IL-6,TNF-αin genioglossus muscle and to explore the correlation between CIH and genioglossus muscle injury,a rat model of SD was established by simulating the pathophysiological characteristics of OSAHS-chronic intermittent hypoxia(CIH).Then,on the basis of CIH,the effects of oxygen therapy on the expression of IL-6,TNF-α in genioglossus muscle of rats were observed,and the effects of oxygen therapy on the expression of IL-6,TNF-α in genioglossus muscle of rats were observed to explore whether oxygen therapy could repair or reverse the genioglossus muscle injury caused by CIH to a certain extent.Methods Preparation of CIH rat model with a self-made plexiglass hypoxia device.Forty healthy male SD rats were randomly and equally divided into normal control group,CIH group,O1 group(oxygen therapy group 1)and O2 group(oxygen therapy group 2)according to the random numerical table method.SD rats were placed in hypoxia chamber and filled with nitrogen and oxygen,each cycle was 120 s,and the gas flow rate was adjusted so that the minimum oxygen concentration in each circulation chamber was about 10% and maintained 45 s.Then gradually restore the oxygen concentration in the cabin to21.YS-1 digital oxygen meter was used to monitor the change of oxygen concentration in the cabin in real time.After 8 hours of intervention every day(9:00-17:00),the animals were fed for 4 weeks,and the model of CIH was completed.Normal control group rats were fed in the normal environment of animal room,all rats were fed with normal water for 4 weeks.Group O1: On the basis of 4 weeks’ CIH,rats were continuously placed in hypoxia chamber,and the oxygen concentration in the chamber was adjusted to about 40%.The rats were fed in the normal environment for the remainder of the day for 4 hours(9:00-13:00),and fed to the 6th week.Group O2: On the basis of the CIH experiment,the rats were kept in hypoxia chamber,the oxygen concentration in the tank was about 50%,and the rats were treated for 4 hours(9:00-13:00)a day.The rats were fed in the normal environment for the remaining period until the 6th week.After feeding the four groups of rats according to the plan,anesthetized rats by intraperitoneal injection of anesthetic,fully exposed genioglossus muscle,removed appropriate size genioglossus muscle tissue and put it into freezing tube,then quickly put it into liquid nitrogen tank,after quick freezing of liquid nitrogen,transfer to-80 ℃ freezer.The effect of CIH on the ultrastructure of genioglossus muscle was observed by transmission electron microscopy(TEM)in control group and CIH group.The histopathological changes of genioglossus muscle in CIH rats were studied by HE staining.Finally,the relative expression of IL-6,TNF-α in genioglossus muscle of each group was detected by Western blot method.Results 1 The weight change of rats before and after the experiment: there was no significant difference in the weight of each group before and after the experiment(P >0.05),and there was no significant difference in the weight of each group after the experiment(P > 0.05).2 Observation on the morphology of genioglossus muscle in rats under transmission electron microscope: the fibers of genioglossus muscle in CIH group were disordered,the fiber spacing was different and the shape of fibers was irregular.In the cells of genioglossus muscle,mitochondria were hypertrophic and swollen,the inner cristae were irregular,some of them were broken,disappeared,and the boundary was unclear.3 HE staining of genioglossus muscle tissue: the shape of genioglossus musclefibers in the control group was regular,arranged neatly,the boundary between themuscle fibers was clear,and the distance between the muscle fibers was approximately equal,in the CIH group,the genioglossus muscle fibers were arranged in disorder and the size of the fibers was different.The fiber is irregular in shape.4 Expression of IL-6 in genioglossus muscle: compared with control group,the expression of IL-6 in genioglossus muscle in CIH group was significantly higher than that in control group(P < 0.05).IL-6,expression in genioglossus muscle in O1 and O2 groups was significantly higher than that in control group(P < 0.05).The expression of TNF-α in genioglossus muscle of O1 and O2 group was higher than that of normal control group(P < 0.05),and the expression of IL-6 in genioglossus muscle of O1 group was significantly higher than that of O2 group(P< 0.05).5 Expression of TNF-α in genioglossus muscle: compared with control group,the expression of TNF-α in genioglossus muscle of CIH group was significantly higher than that of control group(P < 0.05),and the expression of TNF-α in genioglossus muscle of O1 and O2 groups was significantly lower than that of CIH group(P < 0.05).The expression of TNF-α in genioglossus muscle of O1 and O2 group was higher than that of normal control group(P < 0.05),and the expression of TNF-α in genioglossus muscle of O1 group was significantly higher than that of O2 group(P < 0.05).Conclusions 1 CIH could induce the abnormal expression of IL-6,TNF-α in genioglossus muscle.2 The abnormal expression of IL-6,TNF-α in genioglossus muscle can cause genioglossus muscle injury.3 Oxygen therapy could decrease the expression of IL-6,TNF-α in genioglossus muscle to a certain extent.4 The effects of oxygen therapy with different concentration on genioglossus muscle were different.Figure 12;Table 6;Reference 154... |
