| Objectives To investigate the expression and correlation of miR-10 b and HOXD10 in placental tissues of patients with preeclampsia and their significance in the occurrence and development of disease.Methods 1 Sixty-three patients hospitalized in the obstetrics department of Tangshan Gongren Hospital from November 2017 to July 2018 were selected.Among them,29 were in normal pregnancy group,14 were in pre-eclampsia group,and 20 were in severe preeclampsia group.All the patients were delivered by cesarean section.2 The expression of miR-10 b in placenta tissues of normal pregnancy group,preeclampsia group and severe preeclampsia group was detected by RT-PCR;the expression of HOXD10 protein in placenta tissues of normal pregnancy group,preeclampsia group and severe preeclampsia group was detected by Western blot;JEG-3 cells were cultured and miR-10 b mimics were transfected by cell transfection technology.The expression of miR-10 b was overexpressed in JEG-3 cells,and the transfection of JEG-3 cells was detected by RT-PCR.After the cell model was successfully constructed,the expression of HOXD10 mRNA and HOXD10 protein in each group was detected by RT-PCR and Western blot.The proliferation and invasion ability of each group were detected by CCK-8 and scratch test.3 SPSS17.0 software was used for statistical analysis of the experimental data,one-way ANOVA was used for comparison between multiple samples,SNK and LSD were used for comparison between two samples,and bivariate correlation analysis was used for correlation analysis.Results 1 The expression of miR-10 b in placenta tissue of severe preeclampsia group and severe preeclampsia group was lower than that of normal pregnancy group(F=1527.224,P<0.05).The expression of miR-10 b in placenta tissue of severe preeclampsia group was significantly lower than that of preeclampsia group(LSD-t=27.312,P<0.05).The expression of HOXD10 protein in placenta tissue was higher than that in normal pregnancy group(F=277.302,P<0.05),and the expression of HOXD10 protein in severe preeclampsia group was higher than that in preeclampsia group(LSD-t=10.308,P<0.05);the expression of miR-10 b was negatively correlated with that of HOXD10(r=-0.961,P<0.05).2 The expression of has-miR-10b-mimics in JEG-3 cells was detected by RT-PCR.The results showed that the expression level of miR-10 b in JEG-3 cells in experimental group was significantly higher than that in the blank control group,the liposome group and the negative group(F=370.429,P<0.05),but there was no significant difference in the expression of miR-10 b between the blank control group,the liposome group and the negative group(P>0.05).After cell transfection is completed,the expression level of HOXD10 mRNA in the experimental group was significantly lower than that in the blank control group,the liposome group and the negative group(F=208.196,P<0.05),but there was no significant difference in the expression level of HOXD10 mRNA in the blank control group,the liposome group and the negative group(P>0.05).After cell transfection is completed,the expression level of HOXD10 protein in the experimental group cells was significantly lower than that in the blank control group,the liposome group and the negative group(F=105.443,P<0.05),but there was no significant difference among the blank control group,the liposome group and the negative group(P>0.05).After cell transfection is completed,the proliferation ability of the experimental group was significantly higher than that of the blank control group,the liposome group and the negative group(F=1985.701,P<0.05).However,there was no significant difference in proliferation between the blank control group,the liposome group and the negative group(P>0.05);after cell transfection is completed,the invasive ability of the experimental group was significantly higher than that of the blank control group,the liposome group and the negative group(F=112.800,P<0.05),while the invasive ability was poor in the blank control group,the liposome group and the negative group.The difference was not significant(P>0.05).Conclusions 1 HOXD10 is the target gene of miR-10 b in maternal placenta,and they are negatively regulated;2 The expression of miR-10 b in the placenta of patients with preeclampsia decreased significantly,and the expression of HOXD10 protein was significantly increased by negative regulation,which became more and more obvious with the development of the disease;3 Overexpression of miR-10 b in JEG-3 cells can significantly reduce the expression of HOXD10 mRNA and HOXD10 protein and enhance the proliferation and invasion of JEG-3 cells;4 miR-10 b and HOXD10 may become targets for early prevention and diagnosis of preeclampsia.Figure 14;Table 11;Reference 62... |
