| Objective: to explore the relationship between NF-κB and Snail in IUAs,the expression level of NF-κB,Snail,E-cadherin,Vimentin were detected in IUAs patients’ and normal patients’ endometrium,associated cell experiments were also carry out.Method: 34 samples of IUAs patients’ endometrium were collected as experiment group by hysteroscope,and 19 samples of normal patients endometrium were collected in the same way as control group.The expression level of NF-κB,Snail,E-cadherin,Vimentin were detected by qRT-PCR,Western Blot,IHC.Ishikawa cells were pretreated with LPS,then wound healing assay and Western Blot were used to detect cell migration and the expression level of NF-κB,Snail,E-cadherin,Vimentin.Results: the expression level of NF-κB,Snail,Vimentin increased significantly in IUAs group,and the expression of E-cadherin decreased significantly in IUAs group(P<0.05).The migration ability of Ishikawa cells enhanced significantly in LPS group(P<0.05),compared to control group,the expression of NF-κB,Snail,Vimentin were increased,and E-cadherin was decreased(P<0.05).Conclusion: NF-κB may be associated with the development of IUAs through regulation of EMT.From all the results,we can speculate that after endometrium injury,epithelial cells differentiate into fibroblast or myofibroblast cells because of elevation of NF-κB,produce excessive ECM and contribute to the fibrosis of endometrium. |
