Background and purpose Fungi are a general term for a large class of eukaryotic microorganisms,which widely exist in nature and can cause infectious,toxic and hypersensitive diseases in humans.In recent years,with the widespread use of antibiotics,glucocorticoid and immunosuppressants,the incidence of fungal infections is gradually increasing.For the treatment of mycosis,early diagnosis is particularly important.With the development of fungal detection technology,more and more methods are applied to fungal detection,including traditional fungal detection methods,such as direct microscopy,fungal culture and so on.KOH wet tablet method is easy to cause insufficient or excessive dissolution of keratin,resulting in confused background.In recent years,in order to compensate for the disordered background of KOH wet method,fluorescence staining method is more and more used in clinical fungal detection.Fluorescence staining has the advantages of simplicity,rapidity and easy interpretation.Others,such as molecular biology technology,mass spectrometry,serological tests,also play an important role in the diagnosis of mycosis.This paper mainly focuses on the diagnosis of superficial mycosis.By comparing the application of fluorescent staining and KOH wet film in superficial mycosis,we try to find the most suitable method which is simple,convenient,easy to operate and sensitive for the diagnosis of superficial mycosis.Method This study collected 729 patients who underwent fungal direct microscopy in the dermatology fungal laboratory of Shenzhen Second People’s Hospital from June 1,2017 to August 31,2017.Thirteen patients with non-superficial mycosis were excluded and 716 cases were included.The study included 379 male patients and 343 female patients,with a male to female ratio of approximately 1.12:1.In this study,548 patients were treated with fluorescent staining,which was the experimental group;168 patients were treated with KOH wet film method,which was the control group.All patients were superficial mycosis,including tinea manus,tinea pedis,tinea corporis,tinea cturis,tinea versicolor and onychomycosis.Results There were 548 cases in the fluorescent staining group and 309 cases in the positive staining group,the positive rate was 56.4%;While there were 168 cases of KOH wet method group and 50 cases of positive patients,the positive rate was 29.8%.Chi-square test was performed on the results of the two groups with SAS9.1 system.The results showed that there was significant difference between the two groups(χ~2=36.5,P<0.001),and the fluorescence staining method was superior to KOH wet method in the application of superficial mycosis.Conclusion Fluorescence staining is superior to KOH wet method in the diagnosis of superficial mycosis,which provides a rapid,simple and reliable basis for the diagnosis of superficial mycosis.
