In recent years,nano titanium dioxide(TiO2)as a new food additive has been widely used in various foods.A number of studies have shown that nano TiO2 can enter into the biological body through the digestive system,and cause the accumulation of reactive oxygen species(ROS)related substances in the biological body,thus causing different diseases of various metabolic systems.Our group has confirmed in previous studies that oral administration of nano TiO2 can lead to the occurrence of glucose metabolism disorder in adult mice.According to recent epidemiological reports,the incidence of diabetes in children is increasing year by year,and the barrier system of children is weak,and the gastrointestinal function is not yet perfect.So whether the effect of nano TiO2on plasma metabolism of children more serious than that of adults?The issue has not been reported.Therefore,this study conducted a series of studies on whether the effect of nano TiO2 on plasma glucose metabolism in young mice was more serious than that in adult mice.In this study,3-week-old male young cd-1(ICR)mice were administered orally nano TiO2,P25 to establish the experimental animal model.Meanwhile,8-week-old male adult mice of the same strain were used as the experimental control group(young experimental group,YN,50 mg/kg body weight;young control group,YC,equal volume PBS;Adult group,AN,50 mg/kg;Adult control group,AC,equal volume PBS).Firstly,the intake of nano TiO2,body weight and organ coefficient in juvenile mice were detected,and the damage of all viscera in mice were observed.Then,the fasting plasma glucose content of young mice was tracked weekly,and compared with adult mice in the experimental group to determine whether the nano TiO2 had any effect on the plasma glucose metabolism of young mice,and whether the effect was more serious than that of adult mice.Finally,in the case that nano TiO2 can cause plasma metabolism disorder in young mice,the molecular mechanism leading to plasma metabolism disorder in young mice was analyzed to explore the similarities and differences between them and adult mice.The study results showed that mice in the YN group showed the disorder of plasma glucose metabolism after 5 weeks of daily oral intake of nano TiO2,while mice in the AN group showed the disorder of plasma glucose metabolism after 12weeks of oral administration.Relevant kit detection,RT-qPCR,Western Blot and other molecular biological experiments were used to verify the molecular mechanism leading to the disorder of plasma glucose metabolism in YN group and AN group.The experimental results showed that the molecular mechanism leading to the disorder of plasma glucose metabolism in young mice was the same as that in adult mice:the activation of endoplasmic reticulum stress caused by nano TiO2and the accumulation of ROS substances in mice led to the insulin resistance of mice,which eventually led to the disorder of plasma glucose metabolism in mice.The research results of this subject lay a foundation for guiding the production of food and the prevention of diabetes in children.
