Idiopathic pulmonary fibrosis(IPF)is an interstitial pulmonary fibrosis disease with unknown etiology,high incidence,difficulty in diagnosis,and difficulty in treatment.Pathological changes focused on alveolar epithelial cell reduction,alveolar collapse,deformation and residual tissue connections,interstitial cell proliferation,and fibrosis.Modern studies have shown that the mechanism of IPF is related to the epithelial-mesenchymal transition(EMT)hypothesis.EMT can initiate fibrosis at the initial stage,and can also promote collagen deposition,scar change,and fibrosis during the fibrosis stage.form.IPF is attributed to the "pulmonary sputum-pulmonary sputum dualism" in Chinese medicine.Its basic pathogenesis is qi deficiency and blood stasis.This group is committed to intervening in the formation of IPF with Yiqi Huoxue Decoction-Buyang Huanwu Decoction.Object:To observe the effect of Buyang Huanwu Decoction on EMT related indicators in the late course of IPF rats,and to explore whether the process of anti-fibrosis effect of Buyang Huanwu Decoction is related to the EMT hypothesis.Method : 72 SPF SD rats were randomly divided into sham operation group,positive group,high-low and low-dose group and model group of Buyang Huanwu Decoction,12 in each group.The disease model was replicated by classical modeling.In addition to the sham operation group,normal saline was injected,and the other groups were injected with bleomycin physiological saline.The second day of the model was started.Each group of Chinese medicine was given different concentrations of anti-fried Chinese medicine,positive.Prednisone,model group and blank group were given to pure water once a day.On the 28 th day,the tissues were taken out and the pathological changes of lung tissues were observed by HE and Masson staining to judge the degree of fibrosis.The content of TGF-β was detected by ELISA,and Col-I,Col-III,E-cad and western blot were detected by immunohistochemistry.The protein content of α-SMA was detected;the results were statistically analyzed with SPSS 21.0.Result:1.HE and masson staining:Compared with the sham operation group,the model group showed a large number of alveolar epithelial cell hyperplasia,inflammatory cell infiltration,alveolar wall thickening,intracavitarymacrophage infiltration,and light blue staining on the wall.Pathological changes such as increased fiber.Compared with the model group,the lung tissue of the drug-administered group had alveolar epithelial cell hyperplasia,inflammatory cell infiltration,macrophage foci in the cavity,thickened alveolar wall,and light blue stained fibers distributed on the thickened wall.Pulmonary fibrosis scores were all reduced.2.The results of Elisa,immunohistochemistry and WB:(1)TGF-β1:Compared with the sham operation group,the expression of the model group was significantly up-regulated(P<0.01).Compared with the model group,the positive control group was significantly lower(P<0.01),and the TGF-β1 content of each group of Buyang Huanwu Decoction was significantly decreased(P<0.01);(2)Col-I andα-SMA:Compared with the sham operation group,the expression of the model group increased significantly(P<0.01),and the expression of the positive control group decreased compared with the model group(P<0.05).The expression of Col-I in the group of Buyang Huanwu Decoction was significantly decreased(P<0.01),and the expression of α-SMA was decreased(P<0.05).(3)E-cad and Col-III:compared with the sham operation group,model group The expression decreased significantly(P<0.01).Compared with the model group,the positive control group had a significant increase in E-cad(P<0.01),and the Col-III expression was up-regulated(P<0.05).The E-type of Buyang Huanwu Decoction group The cad increased significantly(P<0.01),and the expression of Col-III increased(P<0.05).Conclusion:The effect of Buyang Huanwu Decoction on anti-fibrosis may be related to the EMT hypothesis.
