| Depression is a common complication of brain tumors,and depressive symptoms have become the first clinical manifestations of glioma in clinical diagnosis.Whether brain tumors are associated with depression,whether brain tumors can cause depression,or depression can lead to tumors,these are the main concerns.However,there is still little known about this.This article studies the association between the molecular mechanism of depression and glioma development to find the answer to this question.In this paper,47 different data sets of two platforms,HG-U133 A and HG-U133Plus2,including 367 depression samples and 1726 glioblastoma samples were systematically analyzed by integrating data.Then 463 MDD differentially expressed genes were obtained by linear regression and classical Bayesian method in the limma package of R language,and the differentially expressed genes of 823 GBM were expressed.It is found that there are 27 common differentially expressed genes in MDD and GBM.Among them,LOX,NPY1 R,SHANK3 and VEGFA genes have been found to be associated with MDD and GBM.After obtaining the differentially expressed genes,localized miRNA prediction was performed using the Targetscan tool.At the same time,the protein interaction network was obtained via the STRING online platform,and its significant core gene module was clustered using the MCODE algorithm.Through Cytoscape analysis of the protein interaction network between MDD and GBM,four overlapping key genes were obtained: CXCR4,VEGFA,MGP and GNB5;and CXCR4 and GNB5 genes also existed in the core gene module.At the same time,functional annotation and enrichment analysis were performed through three aspects of differentially expressed genes,significant core gene modules,and predicted miRNAs.The results showed that there were 23 KEGG biologically regulated common pathways in MDD and GBM,and textual mining by COREMINE,a medical ontology information retrievalplatform,identified 13 biological pathways as common biological pathways for the two diseases.Finally,use Apriori algorithm to analyze the differentially expressed genes and GO biological pathways of depression and glioma,and further construct the molecular mechanism network of depression and glioma.The results showed that PLA2G5,LOC644794,VEGFA,MYO10,DAB2 IP,FGFR2,the 6 gene exists in the molecular mechanism network of MDD;PRKCZ,CDC42,EGFR,PAK1,CAMK2 A,CAMK2B,VEGFA,GABRG2,GABRA5,MAL2,HCN1,CPEB3,TAC1,the 13 genes found in the molecular network of GBM.It was also found that the VEGFA gene was significantly associated with the molecular mechanisms of MDD and GBM.Studies have shown that the molecular mechanism of severe depression and glioblastoma is closely related.The common regulation pathway found by functional enrichment analysis will contribute to the pathogenesis of depression and glioma.This study can lay a foundation for biomarkers of depression and glioma,provide new ideas for accurate diagnosis and treatment of diseases,and provide new ways to correctly explain the association between depression and glioma. |
