| Objective:To investigate the potential involvement of LncRNA TCONS00003757 in papillary thyroid carcinoma(PTC),to analyze its relationship with clinical features,and to explore the possible mechanism of action of LncRNA TCONS00003757.Methods:The expression of LncRNA TCONS00003757 was detected by RT-PCR in 60 patients with PTC who underwent in department of head and neck surgery of Tangshan Workers’Hospital from October 2017 to August2018.The clinical data of PTC patients were collected,and the correlation between the expression of LncRNA TCONS00003757 and clinical data was analyzed by χ2 test.Western blot was used to detect the expression differences of ST3GAL5,PI3K,p-Akt and p-mTOR between PTC tissues and normal thyroid tissues.Paired t-test was used to compare the differences between PTC and adjacent normal thyroid tissue.Results:RT-PCR showed that the expression of LncRNA TCONS00003757 was up-regulated in PTC compared to adjacent non-cancerous tissues.χ2 test showed that the expression of LncRNA TCONS00003757 in thyroid carcinoma was significantly correlated with lymph node metastasis and extra-glandular invasion,the difference was statistically significant(P<0.05);and There was no significant relationship between the expression of LncRNA TCONS00003757 and age,sex,tumor size,TMN stage,thyrotropin(TSH),free-triiodo thyronine(FT3),free thyroxine(FT4),thyroglobulin antibodies(Anti-Tg),and thyroid peroxidase antibody(Anti-TPO)(P>0.05).Conclusion:LncRNA TCONS 00003757 is up-regulated during PTC carcinogenesis.The expression of LncRNA TCONS00003757 in PTC is significantly associated with lymph node metastasis and extra-glandular invasion.It may promote the occurrence and migration of PTC by down-regulating ST3GAL5 and then activate PI3K/Akt/PKB signaling pathway.And enhance the tolerance to chemotherapeutic drugs by up-regulating the expression of P-gp protein,affecting the prognosis of PTC.This finding may provide a theoretical basis for the choice of PTC treatment. |
