Objective:Aim to investigate the expression of B7-homology 3(B7-H3)and cytotoxic T lymphocyte-associated antigen-4(CTLA-4)in non-small cell lung cancer and their relationship with clinicopathological characteristics as well as exploring the correlation between B7-H3 and CTLA-4,providing a theoretical basis for the clinical treatment of non-small cell lung cancer.Methods:70 specimens of non-small cell lung cancer(NSCLC)which underwent surgical resection and puncture biopsy(complete record of clinicopathological data)were collected as observation group From January2016 to August 2018 in the Department of Pathology,Petroleum Clinical Medical College of Hebei Medical University,including 48 adenocarcinomas,19 squamous cell carcinomas,3 adenosquamous carcinomas.In addition,30normal lung tissues(more than 5 cm outside the margin of cancer tissue)adjacent to cancer were taken from the above-mentioned 70 cases as control group.Immunohistochemical SP method was used to detect membrane B7-H3and CTLA-4.SPSS 21.0 statistical software was used to analyze the level of B7-H3 and CTLA-4 in the specimens of NSCLC and clinicopathological characteristics,as well as the correlation between B7-H3 and CTLA-4.Results:1 The expression of B7-H3 in non-small cell lung cancerThe expression rate of B7-H3 was 64.2%(45/70)in NSCLC and 6.7%(2/30)in adjacent tissues,it was indicated a statistically significant difference(χ2=27.988,P<0.001).2 The relationship between expression of B7-H3 and clinicopathological featuresThe expression of B7-H3 in smoking group,T3-4 stage group,III-IV stage group,low differentiation group and lymph node metastasis group was significantly higher than that in non-smoking group,T1-2 stage group,I-II stage group,high-medium differentiation group and non-lymph node metastasis group,the differences were statistically significant(χ2=5.877,5.609,18.341,16.593,14.979,P<0.05).The expression of B7-H3 was not correlated with age,sex,maximum diameter of tumors and distant metastasis(P>0.05).3 The expression of CTLA-4 in non-small cell lung cancerThe expression rate of CTLA-4 in NSCLC was 57.1%(40/70),and there was no CTLA-4 expression in adjacent tissues.the difference was statistically significant(χ2=28.571,P<0.001).4TherelationshipbetweenexpressionofCTLA-4and clinicopathological featuresThe expression of CTLA-4 in III-IV stage group,low differentiation group,lymph node metastasis group and distant metastasis group was significantly higher than that in I-II stage group,high-medium differentiation group,non-lymph node metastasis group and non-distant metastasis group,the differences were statistically significant(χ2=21.886,6.076,17.662,7.977,P<0.05).The expression of CTLA-4 was not correlated with age,sex,smoking history,maximum diameter of tumors and T stage(P>0.05).5 The correlation between B7-H3 and CTLA-4The correlation analysis showed that there was a positive correlation between the expression of B7-H3 and CTLA-4 in NSCLC(r=0.740,P<0.05).Conclusion:1 The expression of B7-H3 is closely related to smoking,high malignant degree of tumor tissue,poor T stage,advanced TNM stage and lymph node metastasis.2 The expression of CTLA-4 is closely related to the high malignant degree of tumor tissue,advanced TNM stage,lymph node metastasis and distant metastasis.3 The expression of B7-H3 and CTLA-4 are positively correlated with each other in NSCLC,indicating that they may play a synergistic role in the occurrence and development of NSCLC patients,and providing a theoretical basis for the application of immunotherapy and a combination of immunoblockers in NSCLC.
