Objective To study the therapeutic effect and mechanism of Xiaoyao Powder on peri-menopausal syndrome of liver stagnation.Methods SD rats were divided into blank group,model group,positive control(estradiol)group,Xiaoyao San low dose group,Xiaoyao San high dose group,and receptor antagonistic(ICI182780)group.The depression state of rats was observed by open field,forced swimming and tail suspension experiment.The pathological changes of hippocampus,hypothalamus,uterus,vagina and ovary were observed by HE staining.The ERβin rat hippocampus was detected by immunohistochemistry and Western Blot.Expression of ERαin hypothalamus,uterus,vagina and ovary,kit for detection of DA,NE,E,Glu,GABA and 5-HT in hippocampus and hypothalamus,and detection of DA,NE and E in serum by ELISA kit method,5-HT,ACTH,CORT,LH,FSH and E2 content.Results The results of open field experiment showed that Xiaoyao Powder could significantly increase the total distance of 5 min exercise in rats(P<0.01).The results of forced swimming test showed that Xiaoyao San could significantly reduce the 5 min immobility time of rats.The results of tail suspension experiment showed that Xiaoyao San could be significantly reduced.The cumulative resting time of the rats was 5 min(P<0.01).The results of HE staining showed that the hippocampus cells in the model group were arranged closely,but the number of neurons decreased,the cells became smaller,the nucleus condensed,and the glial cells proliferated.The hippocampus in the Xiaoyao San group were arranged closely,and the number was compared.Many,normal morphology,no obvious nuclear pyknosis;model group rats hypothalamic cell body swelling,membrane rupture,partial atrophy,vacuolar degeneration or necrosis,glial cells significantly proliferated,Xiaoyao San group rat hypothalamus The degree of cell swelling was relieved,and the gliosis of the glial cells was significantly reduced.In the model group,the endometrium of the rats was thin,the secretory vesicles in the epithelium were less,the lamina propria of the lamina propria were less,and the glandular cavity was small.The endometrium of the rats in Xiaoyao San group Thicker,more secretory vesicles in the epithelium,more uterine glands in the lamina propria;periodic changes in the number of vaginal epithelial cells in the model group;no nucleated keratinocytes in the vagina of the Xiaoyao powder group;model group rats The surface epithelium of the ovary is a single-layer squamous cell,and the number of follicles in each stage is reduced,and there is no corpus luteum.In the Xiaoyao San group,the ovary of the ovary is normally developing with more follicles,increasing follicles and degenerating follicle Small,increase in the number of corpora lutea.Immunohistochemistry and Western Blot showed that the expression of ERαin the hippocampus of the model group was significantly decreased in the ERβ,hypothalamus,uterus,vagina and ovary(P<0.01).Xiaoyao San could significantly increase ERβin the hippocampus,hypothalamus,Expression of ERαin uterus,vagina and ovary(P<0.01).The results of ELISA kit showed that the contents of GABA,5-HT,DA,NE and E in the hippocampus and hypothalamus of the model group were significantly increased,and the Glu content was significantly decreased(P<0.01).Xiaoyao San could significantly reduce the large The content of DA,NE,E,GABA and 5-HT in hippocampus and hypothalamus increased the content of Glu(P<0.01).The results of ELISA kit showed that the levels of DA,E,NE,5-HT,ACTH,CORT,FSH and LH in the model group were significantly higher than those in the blank group(P<0.01),E2The content decreased significantly(P<0.01).Xiaoyao San significantly reduced the contents of DA,NE,E,5-HT,ACTH,CORT,FSH and LH in the serum of rats(P<0.01),and significantly increased the content of E2.Conclusions The experimental results show that Xiaoyao Powder has an effect on depression behavior;Xiaoyao Powder can improve the morphology of hippocampus and hypothalamus and ensure its normal physiological functions;Xiaoyao San through DA,NE,E,Glu,GABA,5-HT,ACTH,CORT,LH,FSH,and E2 have a regulatory effect on the hypothalamic-pituitary-ovarian axis,which in turn promotes regulation of the hypothalamic-pituitary-adrenal axis and exerts a peri-menopausal syndrome of the liver-stagnation type(female menopause).The role of depression;after the addition of ICI182780,ERαand ERβare antagonized,indicating that they act through ER receptors. |
